以2%甲酸乙腈溶液为提取液,采用MCX固相萃取,荧光检测器检测,激发波长280nm,发射波长450nm,外标法定量,本项目建立了同时测定饲料中4种氟喹诺酮类药物的反相高效液相色谱分析方法。4种药物在5—500μg/ml浓度范围内呈良好的线性关系,相关系数R。大于0.99996。添加药物水平20--500μg/kg的回收率在75.6%一97.2%。环丙沙星、恩诺沙星、沙拉沙星的定量检出限为20μg/kg,达氟沙星为4μg/kg。试验结果表明:该方法快速简便,准确可靠,重现性好,可用于监测饲料中氟喹诺酮类含量。

A reversed-phase High Performance Liquid Chromatography method was developed for the simultaneous analysis of four fluoroquinolones in animal feed. The determination conditions were as follows: 2% formic acid in acetonitrile solution was used as extraction solvent,MCX solid phase ex- traction was used for extraction, fluorescence detector, excitation wavelength of 280ram, emission wave- length of 450mm,external standard method were used for determination. The linear range for the de- termination of four fluoroquinolones was 5--500μg/ml with R2〉0.9996, showing very good linear rela- tion. The recoveries laid in the range of 75.6%--97.2% at spiking level of 20--500μg/kg. The quanti- tative detection limit is 20μg/kg for ciprofloxacin,enrofloxacin and sarafloxacin and 4μg/kg for danofloxacin. The results show that this determination method is repid and simple,a ccurate and reli- able,good at reproducibility,which can be used to monitor the contents of fluoroquinolones in animal feed.
