为了探索纳米磷酸钙(NCaP)作为猪瘟多肽疫苗佐剂的可行性,本研究利用猪瘟多肽疫苗作为抗原,制备纳米磷酸钙吸附猪瘟多肽疫苗。将制备好的疫苗免疫兔后,分别检测兔抗体水平和攻毒后体温变化情况。检测结果显示,纳米磷酸钙对猪瘟多肽吸附率达70%;免疫后纳米磷酸钙佐剂组抗体水平低于弗氏佐剂,但与游离猪瘟多肽疫苗组相比,有较大提高;除弗氏佐剂组外,所有实验兔均有体温升高现象,纳米磷酸钙佐剂组体温升高的时间与空白组相比推迟16 h。结果表明,NCaP作为佐剂相对游离猪瘟多肽可提高其免疫效果,但其诱导的抗体水平低于弗氏佐剂组。

위료탐색납미린산개(NCaP)작위저온다태역묘좌제적가행성,본연구이용저온다태역묘작위항원,제비납미린산개흡부저온다태역묘。장제비호적역묘면역토후,분별검측토항체수평화공독후체온변화정황。검측결과현시,납미린산개대저온다태흡부솔체70%;면역후납미린산개좌제조항체수평저우불씨좌제,단여유리저온다태역묘조상비,유교대제고;제불씨좌제조외,소유실험토균유체온승고현상,납미린산개좌제조체온승고적시간여공백조상비추지16 h。결과표명,NCaP작위좌제상대유리저온다태가제고기면역효과,단기유도적항체수평저우불씨좌제조。
In order to explore the feasibility of calcium phosphate nanoparticles(NCaP) as a vaccine adjuvant,the present study was conducted using NCaP adsorbed peptide vaccine against classical swine fever(CSF).Rabbits were immunized with the vaccine preparations.Antibody levels in blood samples were detected and body temperatures were measured after rabbits were challenged with CSF spleen viruses.Results showed that a certain amount of CSF peptides were adsorbed on NCaP with the adsorption rate of 70%.Antibody levels of NCaP peptide vaccine group were higher than those of peptide alone group but lower than those of Freund's adjuvant group.The increase of body temperatures postponed 16 hours in NCaP peptide vaccine group as compared with peptide alone group.This study showed that NCaP played an adjuvant effect on CSF peptide vaccine to a certain extent.
