目的:观察低氧和不同运动强度对大鼠血清血管内皮生长因子(VEGF)含量及心肌VEGF基因和蛋白表达的影响。方法:SD大鼠随机分成6组:常氧对照组(NC),低氧对照组(HC),常氧低强度运动组(NEL),低氧低强度运动组(HEL),常氧高强度运动组(NEH),低氧高强度运动组(HEH)。NC、NEL和NEH组在常氧环境下生活,HC、HEL和HEH组在氧含量14.1%的低氧环境下生活。NEL和HEL、NEH和HEH组运动强度分别为15.2 m/min和26.8 m/min,跑台坡度均为10°,40min/天,6天/周。各组大鼠经实验在运动组最后一次运动后24h取血和心脏,检测VEGF含量,运用RT-PCR和WesternBlotting技术检测心肌VEGFmRNA和蛋白表达量。结果:血清VEGF含量随缺氧程度的加深有逐渐下降的趋势,且HEH组下降最为显著(P﹤0.01);RT-PCR和Western Blotting心肌VEGFmRNA和蛋白结果随缺氧程度的加深表达量呈逐渐升高趋势。结论:(1)低氧、运动以及低氧运动都能使血清VEGF含量减少,而此时心肌VEGF基因和蛋白表达均增强。推测是肌组织对循环中VEGF的摄取、利用增加,减少了血清VEGF含量;(2)VEGF的基因和蛋白表达程度与缺氧刺激程度关系密切;(3)缺氧相关因子含量和表达量的变化可作为机体对缺氧适应能力评价的指标。

목적:관찰저양화불동운동강도대대서혈청혈관내피생장인자(VEGF)함량급심기VEGF기인화단백표체적영향。방법:SD대서수궤분성6조:상양대조조(NC),저양대조조(HC),상양저강도운동조(NEL),저양저강도운동조(HEL),상양고강도운동조(NEH),저양고강도운동조(HEH)。NC、NEL화NEH조재상양배경하생활,HC、HEL화HEH조재양함량14.1%적저양배경하생활。NEL화HEL、NEH화HEH조운동강도분별위15.2 m/min화26.8 m/min,포태파도균위10°,40min/천,6천/주。각조대서경실험재운동조최후일차운동후24h취혈화심장,검측VEGF함량,운용RT-PCR화WesternBlotting기술검측심기VEGFmRNA화단백표체량。결과:혈청VEGF함량수결양정도적가심유축점하강적추세,차HEH조하강최위현저(P﹤0.01);RT-PCR화Western Blotting심기VEGFmRNA화단백결과수결양정도적가심표체량정축점승고추세。결론:(1)저양、운동이급저양운동도능사혈청VEGF함량감소,이차시심기VEGF기인화단백표체균증강。추측시기조직대순배중VEGF적섭취、이용증가,감소료혈청VEGF함량;(2)VEGF적기인화단백표체정도여결양자격정도관계밀절;(3)결양상관인자함량화표체량적변화가작위궤체대결양괄응능력평개적지표。
Objective:This study was to observe both effects of hypoxia and exercise on serum vascular endothelium growth factor(VEGF) content and myocardial VEGF gene and protein expressions.Methods:SD rats were randomly assigned to six groups: normoxia control group(NC),hypoxia control group(HC),normoxia low-intensity exercise group(NEL),hypoxia low-intensity exercise group(HEL),normoxia high-intensity exercise group(NEH) and hypoxia high-intensity exercise group(HEH).NC,NEL and NEH were living in normoxia.HC,HEL and HEH were exposed to hypoxia,the oxygen content was 14.1%.NEL and HEL exercise intensity were 15.2m/min,NEH and HEH were 26.8m/min,the slope of treadmill was 10 degree,40min/day,6days/week.After experiment,blood and heart were get after 24h of last training.The content of VEGF was tested.The gene and protein expressions of VEGF were detected by RT-PCR and Western Blotting.Results The content of serum VEGF had descent trend with the deepen of hypoxia level.The result of myocardial VEGFmRNA and protein expressions had gradually increase trend with deepen of hypoxia level.Conclusion(1) Hypoxia,exercise,hypoxia exercise all can make the content of serum VEGF decrease,at the same time the gene and protein expressions of VEGF of myocardial all increase,presume muscular tissue incept and utilize increase of circulation VEGF,decrease serum VEGF content;(2)The gene and protein expressions of VEGF had close relationship with the level of hypoxia stimulate;(3)The variety of content and expression of hypoxia relevant factor could be as the evaluate index of the body to the oxygen deficit adaptive ability.
