目的:研究骨碎补总黄酮对体外培养的人牙周膜细胞(hPDLC)增殖、总蛋白合成及超微结构的影响。方法:运用组织块法,体外培养hPDIJC并进行免疫组织化学鉴定;采用噻唑蓝(MTT)法观察不同浓度骨碎补总黄酮在不同时间作用于体外培养的hPDLC增殖情况;运用BCA法测定不同质量浓度骨碎补总黄酮在不同时间点对hPDLC总蛋白合成的影响;运用透射电镜观察骨碎补总黄酮作用于hPDLC后细胞超微结构的变化。结果:0.1、1、10mg/L质量浓度的骨碎补总黄酮对hPDLC增殖和总蛋白的合成具有促进作用。骨碎补总黄酮作用于人牙周膜细胞后,细胞内粗面内质网、核糖体、线粒体明显增多。结论:骨碎补总黄酮有促进hPDLC增殖的作用,可促进总蛋白的合成,细胞代谢,使功能更活跃。

Objeclive: To investigate the effects of Drynaria total flavonoids on the proliferation ,total protein content and ultra- structure in cultured human periodontal ligament cells( hPDLCs). Methods: hPDLCs were primarily cultured by tissue - culture method in vitro, and identified through imm - unohistichemical method. MTr was used to observe the proliferation of hPDLCs with the different concentrations of Drynaria total flavonoids at different times. BCA Protein Assay Kit was adopted to explore the total protein content of hPDLCs with the effects of different concentration of Drynaria total flavonoids at different times. The ultra- structure of hPDLCs with the effects of Drynaria total flavonoids was observed by Transmission electron microscope. Results: Drynaria total flavonoids at dose of 0. 1,1,10mg/L in cultures resulted in an increase of total protein content. There were more rough endoplasmic reticulum, ribosome and in the experimental group than in the control. Conclusion: Drynaria total flavonoids may stimulate the proliferation,total protein synthesis and metabolism of hPDLCs.
