建立同时测定秦艽中龙胆苦苷和牛膝中蜕皮甾酮含量的RP—HPLC分析方法。采用KromacilC18色谱柱(4.6mm×250mm,5μm);流动相:乙腈(A)-0.1%醋酸水溶液(B)进行梯度洗脱;流速1.0mL·min^-1;检测波长:254nm;柱温:25℃。龙胆苦苷、蜕皮甾酮的进样浓度分别在0.80~20.00,0.40~10.00μg·mL^-1。范围内线性关系良好;平均回收率(n=6)分别为100.02%,99.95%。结论:该方法简便,准确,重现性好,具有较强的应用价值。

To develop an HPLC method for determining the content of gentiopicrin and ecdysterone in Gentiana macrophyUa Pall and Achyranthes bidentata B1.A Kromacil Cls column (4.6mm×250mm,5μm) was adopted,the mobile phase was acetonitrile-water (contained 0.1% acetic acid) for gradient elution with flow rate of 1.0 mL·min^-1.The detection was set at 254 nm,and the column temperature was maintained at 25 ℃.Good linearities were obtained from 0.80 to 20.00,0.40 to 10.00 μg·mL^-1 for gentiopicrin and ecdysterone,respectively. The mean recoveries (n=6) of 2 components were 100.02%,99.95%,respectively. Conclusion: The method is simple and accurate with good reproducibility.
