[目的]本试验的目的是优化秦川牛瘤胃微生物DNA提取方法。[方法]针对秦川牛特有的生理及生物学特性,采取珠磨法、反复冻融法、液氮研磨法、超声波法提取微生物DNA,并获得一种提取秦川牛瘤胃胃微生物DNA的最佳方法。[结果]珠磨法、反复冻融法提取的DNA片段较完整,DNA含量较高。[结论]珠磨法和反复冻融法是较科学的提取方法。

[목적]본시험적목적시우화진천우류위미생물DNA제취방법。[방법]침대진천우특유적생리급생물학특성,채취주마법、반복동융법、액담연마법、초성파법제취미생물DNA,병획득일충제취진천우류위위미생물DNA적최가방법。[결과]주마법、반복동융법제취적DNA편단교완정,DNA함량교고。[결론]주마법화반복동융법시교과학적제취방법。
[Objective]The purpose was to optimize DNA extraction method from rumen microorgan isms in Qinquan cattle. [Method]The bead mill, repeated freezing and thawing method, liquid nitrogen, grinding, ultrasonic were taken to extract microbial DNA according the unique physiological and biological characteristics of Qinchuan cattle. [Result]The result showed that the integrity and content of DNA fragments were better by bead mill, repeated freezing and thawing methods. [Conclusion]The results showed that the selected bead mill and repeated freeze thaw methods were better method to extract DNA from rumen microorganisms in Qinquan cattle.
