利用筛选和验证相结合的方法筛选出了产碱性脂肪酶活性较高的菌株BL1011。经过形态观察、生理生化试验及分子生物学鉴定,结果表明该菌株为假单胞菌(Pseudomonas sp.)。运用单因素试验和均匀试验优化了BL1011菌株摇瓶产酶培养基和最佳发酵条件。在培养基为麦芽糖2.5%,蛋白胨3.0%,大豆油0.5%,K2HPO40.2%,培养条件为起始pH值7.5,温度33℃,转速180r/min,装液量20mL,发酵周期为60h的条件下,酶活力达到最高,为223U/mL。

An alkaline lipase-producing strain BL1011 was screened quickly through the screening and confirmation.According to morphologic observation ,physiology biochemical experiment and molecular identification ,this strain was Pseuclornonas sp.Using single factor and uniform design approaches, the best fermentation medium and fermentation condition were optimized. The optimized culture medium was maltose 2.5%, peptone 3.0%, soya-bean oil 0.5% ,K2HPO4 0.2% .The initiative pH,temperature,agitation speed and medium volume were 7.5,33 ℃ ,180 dmin and 20 mL, respectively. The highest enzyme activity was 223 U/mL after 60 hours.
