在酸性条件下,以4一羰基苯甲醛为衍生化试剂分别与4种硝基呋喃类代谢物(AOZ、SEM、AMOZ和AHD)反应制成相应丰抗原:CPAOZ、CPSEM、CPAMOZ和CPAHD.采用碳二亚胺法将半抗原与牛血清蛋白偶联合成人工抗原:CPAOZ—BSA、CPSEM—BSA、CPAMOZ—BSA和CPAHD—BSA,产物经紫外扫描鉴定,表明抗原偶联牛血清蛋白成功。在此基础上研制成4种硝基呋喃代谢物快速胶体金试剂条,最低检出限分别为O.5、1.0、1.0、2.0μg/kg,相互间的交叉反应率小于0.1%。

With 4-carbonyl benzaldehyde for derivatization reagent, four nitrofuran metabolites (AOZ, SEM, AMOZ and AHD)were made into the corresponding haptens:CPAOZ,CPSEM,CPAMOZ and CPAHD in the acidic conditions,and the haptens were successfully conjugated to bovine serum albumin (BSA)with the carbodiimide method to form artificial antigens, such as CPAOZ- BSA, CPSEM-BSA, CPAMOZ-BSA and CPAHD- BSA. Products by UV scanning showed success of antigens linked into BSA. On the basis of this study,colloidal gold rapid test devices for nitrofuran metabolites were successfully developed. ,The minimum detection limits were 0.5 μg/kg, 1.0 μg/kg, 1.0 μg/kg and 2.0 μg/kg respectively, and mutual cross reaction rate was less than 0.1%.
