本文通过提取、分离、鉴定连翘(Forsythia suspensa)中酪氨酸酶抑制成分,并检测其酪氨酸酶抑制活性.采用的方法是利用蘑菇酪氨酸酶对连翘进行抑制活性筛选,并通过硅胶柱色谱法分离、鉴定酪氨酸酶抑制成分.结果发现,对连翘的乙醇提取物及其二氯甲烷、正丁醇、水萃取物进行酪氨酸酶抑制活性检测结果均有浓度依赖性的抑制活性,尤其是连翘的二氯甲烷萃取物显示最强的活性,并通过硅胶柱色谱法分离得到有效成分,利用核磁共振、质谱等分析手段有效成分被鉴定为白桦脂酸,其酪氨酸酶的半数抑制率IC50值为138.5±1.57μmol/L.结论:连翘具有较强的酪氨酸酶抑制活性,为连翘的有效利用提供实验依据.

Objective: Extraction, Isolation and Identification of tyrosinase inhibitory active ;i constituent From Forsythia suspensa. Method: The fruits of F. suspensa were screened by mushroom tyrosin'ase, and the tyrosinase inhibitory active constituent was isolated via silica gel chromatography and identified by NMR and Mass spectra. Result: The ethanol extract, and fractions of CH2C12, BuOH and H2O from F. suspensa showed the tyrosianse inhibitory activity in a dose-dependent manner. In particular, the CH2 C12 fraction appeared the most activity. The tyrosinase inhibitory active constituent was isolated by silica gel chromatography and identified as betulinic acid, showing the ICs0 value of 138.5 ±1.57μmol/L. Conclusion: F. suspensa showed strong tyrosinase inhibitory activity, and it was provided for the effective use of F. suspensa.
