应用放射免疫分析法(CharmⅡ)检测动物组织及尿样中残留四环素类药物,对猪的组织及尿样进行了金霉素标准品添加验证试验.选取6个样品进行添加并时每个样品检测3次,结果显示猪尿样在加标点1000μg/L,稀释因子50的条件下,其检测值在同一样品中变异系数0.50%~2.63%,样品间变异系数5.05%;猪的组织(猪瘦肉)在加标点100μg/kg,稀释因子4的条件下,其检测值在同一样品中变异系数0.49%~7.44%,样品间变异系数18.33%.对猪尿样降低稀释因子由说明书要求的150降到50,结果表明在稀释因子50的条件下,增大了的样本背景干扰对试剂盒检测基本无影响.应用该方法进行四环素类药物残留的大类筛选,其结果检测稳定性良好.

Charm Ⅱ radioimmunoassay was established for the total-tetracyclines residues in pork and urine. We have made verification tests to add the staiadard solution of chlorotetracycline in negative residues samples of pork and urine, selected 6 samples of pork and urine, respectively, and detected 3 times for each sample. For added samples of urine, the coefficient of variation (CV) of intra-group is between 0.50% to 2.63%, the CV of inter-group is 5.05 % at punctuate 1 000μg/L and dilution factor as 50 For added samples of pork, the CV of intra-group is between 0.49% to 7.44 %, the CV of inter-group is 18. 33 % at punctuate 100 μg/kg and dilution factor as 4. Its result suggests that therers almost no affection for the lower dilution factor from 150 to 50 which increases the background interference. The results of confirmatory tests suggest that the radioimmunoassay method can be used for the screening of tetracyclines residues in pork and urine.
