目的:观察夏枯草提取物对人食管癌Eca-109细胞增殖和凋亡的影响,探讨夏枯草抗肿瘤作用及机制。方法:不同浓度夏枯草作用于食管癌Eca-109细胞,利用MTr法检测细胞增殖抑制率,免疫组织化学法检测Bcl-2、Bax蛋白表达;Annexin—V—FITC/PI法检测细胞凋亡率。结果:中高剂量夏枯草抑制食管癌Eca-109细胞增殖,并呈量效和时效关系;使细胞凋亡率增高,同时Bcl-2蛋白减少,Bax蛋白表达增加。结论:夏枯草提取物可抑制食管癌Eca-109细胞增殖,使Bcl-2蛋白表达减少,Bax蛋白表达增加,细胞凋亡增加。

목적:관찰하고초제취물대인식관암Eca-109세포증식화조망적영향,탐토하고초항종류작용급궤제。방법:불동농도하고초작용우식관암Eca-109세포,이용MTr법검측세포증식억제솔,면역조직화학법검측Bcl-2、Bax단백표체;Annexin—V—FITC/PI법검측세포조망솔。결과:중고제량하고초억제식관암Eca-109세포증식,병정량효화시효관계;사세포조망솔증고,동시Bcl-2단백감소,Bax단백표체증가。결론:하고초제취물가억제식관암Eca-109세포증식,사Bcl-2단백표체감소,Bax단백표체증가,세포조망증가。
[Objectivel To investigate the effect of Prunella vulgaris on esophageal cancer Eca-109 cells proliferation and apoptosis. [Method] Esophageal cancer Eca-109 cells were co-cultured with various of Prunella vulgaris for 24, 48, 72h respectively. The rate of cell proliferation inhibition was detected by MTY assay. The levels of bax and Bel-2 expression were detected by immunohistochemistry. |Methodl The cells apoptosis ratio were detected by Annexin-V-FITC/PI kit. [Result] Prunella vulgaris could effectively inhibit esophage- al cancer Eca-109 cells proliferation in dose-dependent and time-dependent manner, increase the level of bax expression and reduce the level of Bcl-2 expression, increase cell apoptosis. [Conclusion] Prunella vulgaris could substantially inhibite esophageal cancer Eca-109 cells proliferation, increase the level of Bax expression, reduce the level of Bcl-2 expression, and induced cell apoptosis.
