本研究从已知感染蜜蜂黑王台病毒(Blackqueencellvirus;BQCV)和蜜蜂畸翅病毒(DeformedWingVirus,DWV)的东方蜜蜂蜂群中采集工蜂个体,将其纵向剖成3份。一份存于-79℃,一份加入1.2mL无水乙醇保存,第三份加入1.2mLTRIzol保存。对3组样本每隔3d同时进行RNA提取,并使用RT—PCR方法检测蜜蜂黑王台病毒和蜜蜂畸翅病毒。结果表明在TRIzol中保存的样品和-79℃冰冻保存的样本所提取RNA相对完整,病毒检测结果一致;在无水乙醇中保存的样品RNA质量相对较差,病毒检测阳性率低于前两者。该研究为蜜蜂病毒检测的采样保存方法提供一定的理论依据。

본연구종이지감염밀봉흑왕태병독(Blackqueencellvirus;BQCV)화밀봉기시병독(DeformedWingVirus,DWV)적동방밀봉봉군중채집공봉개체,장기종향부성3빈。일빈존우-79℃,일빈가입1.2mL무수을순보존,제삼빈가입1.2mLTRIzol보존。대3조양본매격3d동시진행RNA제취,병사용RT—PCR방법검측밀봉흑왕태병독화밀봉기시병독。결과표명재TRIzol중보존적양품화-79℃빙동보존적양본소제취RNA상대완정,병독검측결과일치;재무수을순중보존적양품RNA질량상대교차,병독검측양성솔저우전량자。해연구위밀봉병독검측적채양보존방법제공일정적이론의거。
For the present study, samples of bee worker were collected from the Apis terra colonies infected with black queen cell virus (BQCV) and deformed wing virus (DWV). Each sample was sliced into three pieces and one of which was stored at -79℃, the other two were kept in 1.2mL 100% ethanol and 1.2mLTRIzol respectively. Aiming at these three different storage methods, RNA extractions and detections of BQCV and DWV using RT-PCR were carried out with each piece at an interval of three days. Analysis of the results indicating that the integrity of RNA extracted from the pieces stored at -79℃ and in TRIzol was rela- tively in good condition while it was not so good for the piece kept in 100% ethanol. Detections of viruses exhibit-ed the same trend.
