研究了利用胰酶粉提取胰激肽释放酶的动态提取工艺。分别考察了醋酸钠添加与否、洗脱的速率、不同高径比的层析柱,梯度洗脱与直接洗脱,一次梯度洗脱与二次梯度洗脱等因素对利用胰酶粉提取胰激肽释放酶的影响。利用胰酶粉提取胰激肽释放酶的动态洗脱胰激肽释放酶的最佳条件为:控制流速为2 mL/min,添加醋酸钠,梯度洗脱及一次洗脱。本实验考察的动态条件为进一步胰激肽释放酶的分离纯化提供了理论依据。

The dynamic extraction experiment of kallikein from trypsogen power was studied.The addition of CH3COONa,the rate of elution,different height to diameter ratio of column chromatography,gradient elution or straight elution,the time of gradient elution on the use of trypsogen power orthogonal static extraction of pancreatic kallikein were investigated.The optimal dynamic kallikein extracted conditions were the flow rate controlled at 2 mL/min,added CH3COONa,and one time of gradient elution.The effect of dynamic conditions was to provide a theoretical basis for separation and purification of kallikein.
