目的:探讨Th17细胞及白介素-17(IL-17)在系统性红斑狼疮(SLE)发病中的作用。方法以SLE患者及健康志愿者为研究对象,流式细胞术检测外周血Th17细胞比例,ELISA法检测血清IL-17浓度,并与SLE疾病活动指数(SLEDAI)进行相关分析。结果①SLE患者外周血Th17细胞占T细胞比例为(2.51±0.76)%,健康对照组Th17细胞比例为(0.68±0.23)%,两组比较差异具有统计学意义(t=17.2,P<0.01);②SLE患者血清IL-17浓度为(114.2±17.1)pg/ml,健康志愿者血清IL-17浓度为(52.2±6.9)pg/ml,两组比较差异具有统计学意义(t=24.3,P<0.01);③SLE患者SLEDAI评分与外周血Th17细胞比例(r=0.74, P<0.01)及血清IL-17浓度(r=0.71, P<0.01)呈正相关。结论 Th17细胞及IL-17可能在SLE发病过程中发挥作用,以二者为靶向的治疗具有应用前景。

목적:탐토Th17세포급백개소-17(IL-17)재계통성홍반랑창(SLE)발병중적작용。방법이SLE환자급건강지원자위연구대상,류식세포술검측외주혈Th17세포비례,ELISA법검측혈청IL-17농도,병여SLE질병활동지수(SLEDAI)진행상관분석。결과①SLE환자외주혈Th17세포점T세포비례위(2.51±0.76)%,건강대조조Th17세포비례위(0.68±0.23)%,량조비교차이구유통계학의의(t=17.2,P<0.01);②SLE환자혈청IL-17농도위(114.2±17.1)pg/ml,건강지원자혈청IL-17농도위(52.2±6.9)pg/ml,량조비교차이구유통계학의의(t=24.3,P<0.01);③SLE환자SLEDAI평분여외주혈Th17세포비례(r=0.74, P<0.01)급혈청IL-17농도(r=0.71, P<0.01)정정상관。결론 Th17세포급IL-17가능재SLE발병과정중발휘작용,이이자위파향적치료구유응용전경。
Objective We attempted to study the role of Th17 cells and interleukin 17 (IL-17) in the pathogenesis of systemic lupus erythematosus (SLE).MethodsA total of 60 SLE patients and 30 healthy subjects were recruited in this study. The frequency of Th17 cells was analyzed by flow cytometry and the serum IL-17 concentration was determined by ELISA. The correlation between SLE disease activity index (SLEDAI) and the frequency of Th17 cells or the serum IL-17 concentration were analyzed.ResultsSLE patients had a higher frequency of Th17 cells in peripheral blood T cells compared with healthy subjects [(2.51±0.76)% in SLE patients vs (0.68±0.23)% in healthy subjects,P<0.01]. Moreover, the serum IL-17 concentration in SLE patients increased more significantly than that in healthy subjects [(114.2±17.1)pg/ml in SLE patients and(52.2±6.9)pg/ml in healthy subjects,P<0.01 respectively]. There was a strong positive association between SLEDAI and the frequency of Th17 cells or the serum IL-17 concentration in SLE patients.ConclusionTh17 cells and IL-17 may play a role in the pathogensis of SLE. Targeting Th17 cells or IL-17 may have a theropeutic potential in the treatment of SLE.
