目的 观察褪黑素(MT)对睡眠剥夺(SD)大鼠空间学习记忆能力的影响.方法 小平台水环境法制备SD大鼠模型.高效液相法检测不同时程SD大鼠松果体内MT含量.Morris水迷宫方法检测大鼠的空间学习记忆能力,每天训练前30 min腹腔注射MT.结果 松果体内MT含量与SD时间有关,随着SD时间的延长,MT含量逐渐下降,睡眠恢复后,MT含量明显增加.与对照组大鼠比较,SD大鼠的逃避潜伏期明显延长,跨越原平台象限次数减少;在SD期间补充外源性MT后,上述指标与对照组比较差异无统计学意义(P>0.05).结论 SD引起的空间学习记忆能力下降与松果体内MT分泌减少有关,补充外源性MT能预防这种因睡眠不足导致的学习记忆能力下降.

Objective To observe the effect of melatonin (MT) on the spatial learning and memory of sleep deprived (SD) rats.Methods The deprivation of sleep was accomplished by maintaining rats on a small platform surrounded by water.High performance liquid chromatography was used to test the MT content in the pineal gland.The spatial learning and memory ability of the rats was tested with Morris water maze.Rats were intraplantarly injected with MT (15 mg/kg),then Morris water maze test was examined 30 min later.Results In the present study,high performance liquid chromatography showed that the MT content in the pineal gland was significantly reduced with the extention of SD,and it was markedly increased after sleep recovery.Compared with control group,the escape latency was prolonged and the numbers of crossing hidden platform were decreased of SD group; and there was no significant difference between control group and SD + MT group.Conclusion The results demonstrate that the reduction of spatial learning and memory ability during SD is related to decreased MT in the pineal gland,and it can be prevented by exogenous MT supply.
