目的:研究短时间吸入七氟醚对发育中大鼠海马神经元凋亡的影响。方法78只1个月龄 SD大鼠按随机数字表法随机分为3组,空气对照组(Sa,26只),载气对照组(Sc,26只),3%七氟醚吸入2 h 组(S2,26只),建立七氟醚吸入模型,吸入结束后12 h 用旷场实验和 T 迷宫进行神经行为学测试,同时断头取海马组织,苏木素-伊红染色观察海马神经元形态学改变,用流式细胞仪检测凋亡,激光共聚焦显微镜测神经元细胞内游离钙离子浓度。结果 S2组海马凋亡神经元数目以及神经元内游离钙离子相对荧光像素值均高于对照组,差异有统计学意义(P<0.05);但神经行为学检测无明显差别。结论七氟醚可诱导幼龄大鼠海马神经元凋亡,可能通过升高细胞内钙离子浓度而触发凋亡。但单次、短时间吸入(<2 h)七氟醚引起的神经元凋亡无明显神经行为学意义。

Objective To study the effects of short-term inhalation of sevoflurane on the hippocampal neuronal apoptosis of rats during puberty. Methods Seventy-eight Sprague-Dawley (SD) rats aged 1 month were randomly as-signed to 3 groups: air control group (Sa, n=26), carrier gas control group (Sc, n=26), 2 h inhalation of 3% sevoflurane group (S2, n=26). The inhalation of sevoflurane model was established. At 12 h after inhalation, all groups underwent neuroethology asessment using open-field and T-maze test. This was followed by decapitation for extraction of the hip-pocampus. The changes of cell morphology in hippocampal neuron were observed via HE staining. Apoptotic cells were measured by flow cytometry, and laser scanning, and confocal microscope was used to measure the concentration of intracellular calcium in the neuron. Results The number of apoptotic neuron, intracellular free calcium concentra-tion and fluorescence intensity in group S2 were significantly increased compared with control groups (all P<0.05). Conclusion Sevoflurane elicits hippocampal neuron apoptosis in rat infants, possibly via increasing the concentration of intracellular calcium. However, neuron apoptosis caused by a single short-time inhalation of sevoflurane (<2 h) did not markedly affect the neuroethology.
