以博落回叶片为外植体材料,在MS培养基的基础上添加不同的激素进行愈伤组织诱导试验,并采用液相色谱-质谱法检测愈伤组织中的血根碱含量,比较不同愈伤组织中的血根碱代谢累积差异。结果表明:Y-9培养基(添加0.75 mg/L 6-BA和0.5 mg/L NAA)上所生长的愈伤组织中血根碱含量最高,达到0.1910 mg/mL;添加NAA的Y-9培养基中的愈伤组织血根碱代谢累积水平较高,为XY-1培养基中愈伤组织血根碱含量的18倍以上,也明显高于其他培养基中愈伤组织血根碱的含量;高浓度的2,4-D不利于血根碱的代谢累积。

Taking the leaf of Macleaya cordata as explant material and adding different hormones to the MS culture medium to conduct callus inducement experiment, and then using UPLC-QTOF to detect the sanguinarine content in callus in order to compare the difference in metabolizing and accumulation of sanguinarine between different calluses. The results showed that the callus grew in Y-9 medium (added with 0.75 mg/L of 6-BA and 0.5 mg/L of NAA) had the highest sanguinarine content, which reached 0.191 0 mg/mL; the metabolizing and accumulation level of sanguinarine in callus grew in Y-9 medium which added with NAA was relatively higher, which was 18-fold of that in callus grew in XY-1 medium, and it also significantly higher than that in callus grew in the other media; high content of 2,4-D was not beneficial for metabolizing and accumulation of sanguinarine.
