本文对里氏木霉产纤维素酶发酵培养条件及对其所产纤维素酶的酶学性质进行初步研究。结果表明,里氏木霉发酵产酶的最佳培养基为:麦麸1.8%、硝酸钠1.3%、碳酸钙0.3%、氯化钠0.2%、磷酸二氢钾0.3%;里氏木霉所产纤维素酶的最适反应条件为:pH 4.0、50℃,金属离子Fe2+、Co2+、Mn2+、Ca2+对酶活有促进作用,而Fe3+、Ag+对酶有抑制作用。经培养基优化后,发酵液上清中的最终酶活为116.64U/mL,是优化前的3.5倍。
本文對裏氏木黴產纖維素酶髮酵培養條件及對其所產纖維素酶的酶學性質進行初步研究。結果錶明,裏氏木黴髮酵產酶的最佳培養基為:麥麩1.8%、硝痠鈉1.3%、碳痠鈣0.3%、氯化鈉0.2%、燐痠二氫鉀0.3%;裏氏木黴所產纖維素酶的最適反應條件為:pH 4.0、50℃,金屬離子Fe2+、Co2+、Mn2+、Ca2+對酶活有促進作用,而Fe3+、Ag+對酶有抑製作用。經培養基優化後,髮酵液上清中的最終酶活為116.64U/mL,是優化前的3.5倍。
본문대리씨목매산섬유소매발효배양조건급대기소산섬유소매적매학성질진행초보연구。결과표명,리씨목매발효산매적최가배양기위:맥부1.8%、초산납1.3%、탄산개0.3%、록화납0.2%、린산이경갑0.3%;리씨목매소산섬유소매적최괄반응조건위:pH 4.0、50℃,금속리자Fe2+、Co2+、Mn2+、Ca2+대매활유촉진작용,이Fe3+、Ag+대매유억제작용。경배양기우화후,발효액상청중적최종매활위116.64U/mL,시우화전적3.5배。
In order to improve the cellulose enzyme production of Trichoderma reesei, culture medium was opti-mized in our research, and the enzymatic properties were studied in this study also. The results of experients showed that the primum culture medium were consisted with 1.8% wheat bran, 1.3% sodium nitrate, 0.3% calcium carbonat, 0.2% sodium chloride and 0.3% potassium dihydrogen phosphate. The optimal reaction conditions of the cellulase is 50℃,pH 4.0. Results also showed that the cellulose activited by some metalions such as Fe2+, Co2+, Mn2+ and Ca2+, inhibited by Fe3+, Ag+ significantly on the country. Under the optimum conditions, the cellusose activity of the ferment brum of Tric hoderma reesei was 3.5 times as before.
