目的:探讨胱硫醚-γ-裂解酶( cystathionine-γ-lyase,CSE)在过敏性休克小鼠心肌组织中的表达和意义。方法:通过注射卵蛋白( OVA)制作小鼠过敏性休克模型。将昆明种小鼠60只随机分为4组,即:PBS对照组、OVA模型组、OVA模型﹢炔丙基甘氨酸( PPG)组、OVA模型﹢硫氢化钠( NahS)组。各组小鼠致敏第3周诱发过敏性休克致死,未死亡小鼠1 h后颈椎脱臼处死动物。分别于死亡后即时(T1)、死亡后冷冻2 d(T2)和冷冻7 d(T3)三个时间点取心肌组织行免疫组织化学染色检测心肌组织CSE蛋白的表达。结果:对照组小鼠心肌组织中CSE呈弱阳性表达,OVA诱导的过敏性休克模型心肌组织中CSE呈阳性表达。给予外源性NahS后CSE在心肌组织中表达增强,而给予h2 S抑制剂PPG后,CSE在心肌组织中表达减弱较为明显。各组小鼠心肌组织在T1、T2、T3三个时间点CSE的表达变化不明显。结论:过敏性休克小鼠心肌细胞中CSE的表达增强,应用免疫组化染色检查心肌组织中CSE的表达可为过敏性休克死亡的法医学鉴定提供形态学参考指标。

목적:탐토광류미-γ-렬해매( cystathionine-γ-lyase,CSE)재과민성휴극소서심기조직중적표체화의의。방법:통과주사란단백( OVA)제작소서과민성휴극모형。장곤명충소서60지수궤분위4조,즉:PBS대조조、OVA모형조、OVA모형﹢결병기감안산( PPG)조、OVA모형﹢류경화납( NahS)조。각조소서치민제3주유발과민성휴극치사,미사망소서1 h후경추탈구처사동물。분별우사망후즉시(T1)、사망후냉동2 d(T2)화냉동7 d(T3)삼개시간점취심기조직행면역조직화학염색검측심기조직CSE단백적표체。결과:대조조소서심기조직중CSE정약양성표체,OVA유도적과민성휴극모형심기조직중CSE정양성표체。급여외원성NahS후CSE재심기조직중표체증강,이급여h2 S억제제PPG후,CSE재심기조직중표체감약교위명현。각조소서심기조직재T1、T2、T3삼개시간점CSE적표체변화불명현。결론:과민성휴극소서심기세포중CSE적표체증강,응용면역조화염색검사심기조직중CSE적표체가위과민성휴극사망적법의학감정제공형태학삼고지표。
Objective:To study the expression and significance of cystathionine-γ-lyase( CSE)in myocardial tissues of mice died of anaphylactic shock. Methods:The model of anaphylactic shock was established by injection of ovalbumin ( OVA). 60 Kunming mice were randomly divided into four groups:PBS control group,OVA model group,OVA model﹢propargylglycine(PPG)group,OVA model﹢sodium hydrosulfide(NahS)group. three weeks after sensitization,OVA was injected into the Tail vein of the mice to induce anaphylactic shock and death,or sacrificed by cervical dislocation. The expressions of cystathionine-γ-lyase in myocardial tissues were examined at immediate( T1 ),frozen( -20 ℃)2 d (T2)and frozen( -20 ℃)7 d(T3)after death by immunohistochemical staining methods. Results:There were weakly positive expressions in myocardial tissues of CSE in control group,and positive expressions of CSE in myocardial tissues induced by OVA in anaphylactic shock model. the expression of CSE increased in myocardial tissue after administration of exogenous NahS,while the expressions of CSE were decreased obviously in the myocardial tissue,after giving h2 S inhib-itors PPG. Changes in the expressions of CSE were not evident in each group at T1,T2,T3. Conclusion:Cystathionine-γ-lyase were expressed in myocardial tissues of anaphylactic shock mice,and the detection of the expression of cystathi-onine-γ-lyase in myocardial tissues of mice died of anaphylactic shock by immunohistochemical staining method may pro-vide an objective index for pathological morphological reference evidences in forensic identification.
