采用徒手切片和显微测量法观察分析铁皮石斛、细茎石斛和齿辨石斛3种药用石斛的显微特征;用傅里叶红外光谱(FT-IR)及其导数光谱法测定其红外光谱特征;用 AFLP 分子标记方法分析其分子鉴别特征.结果表明,3种药用石斛显微特征存在一定差异,铁皮石斛的角质层比较薄,厚度约4.9μm;3种药用石斛茎表皮细胞为不均匀加厚型、无增厚型和均匀加厚型,齿瓣石斛的厚壁组织细胞壁加厚程度明显,厚度为30.0μm.在3种药用石斛二阶导数光谱特征中,1585、1217和1049 cm -1是细茎石斛的特有峰,1569、1450、1225和1073 cm -1是铁皮石斛和齿瓣石斛的共有特征峰,1011 cm -1是细茎石斛和齿瓣石斛的共有特征峰. AFLP 扩增和聚类分析结果表明,3种药用石斛的总扩增位点为113个,其中多态性扩增位点共111个,铁皮石斛特异性扩增位点明显多于细茎石斛和齿瓣石斛.显微结构观察、红外光谱和 AFLP 分子鉴定方法的综合应用对3种药用石斛提供了更准确的鉴别信息.

The freehand section and micrometer analysis were conducted to study the microscopic characters of the stems of Dendrobium officinale,D. moniliforme and D. devonianumt. The IR characters of three medicinal Dendrobii were analyzed by FT-IR and the second order derivative FT-IR. The molecular characters of the three medicinal Dendrobii were amplified and sequenced by AFLP. Results showed that the cell wall thickness of cuticula of D. offi-cinale was 4. 9 μm that was fairly thinner than the other two species. The cell wall thickness of stem epidermis in D. officinale is 9. 1 μm,with non uniform thickness,D. moniliforme has non thickened,and D. devonianum has uniform thickened. The characteristic peaks in the second order derivative FT-IR for D. moniliforme are 1 585, 1 217 and 1 049 cm - 1 ,D. officinale and D. devonianum have special peaks of 1 569,1 450,1 225 and 1 073 cm - 1 ,D. moniliforme and D. devonianum had the special peak of 1 011 cm - 1 . The three medicinal Dendrobii gen-erated 113 amplified loci,111 of which was polymorphic loci which accounted for 99. 7% ,and the special loci number of D. officinale was obviously more than other two species in the total amplified loci by AFLP. The three medicinal Dendrobii can be distinguished by the comprehensive application of the microscopic,IR and AFLP char-acteristics.
