目的:探讨小柴胡汤对 C6胶质瘤大鼠 ESM-1和 VEGF 蛋白表达水平的影响。方法清洁级 Wistar 老年雄性大鼠18月龄,n=60只,右前肢腋窝皮下接种2×106/20μl 的细胞悬液;然后随机分为6组:肿瘤对照组、阳性药A 组(替莫唑胺;50 mg/Kg)、阳性药 B 组(替莫唑胺;25 mg/Kg)、小柴胡汤 C 组(50 g/Kg)、小柴胡汤 D 组(20 g/Kg)、小柴胡汤 E 组(10 g/Kg);每组均分别灌胃给予生理盐水、阳性药 A、阳性药 B、小柴胡汤 C、小柴胡汤 D 和小柴胡汤E,连续服药5 d,休息4 d,共给药3次,于最后一次给药后2 d 处死动物,眼球取血及取肿瘤组织。采用 ELISA 酶联免疫法检测大鼠血清及肿瘤组织中 ESM-1和 VEGF 的蛋白含量;采用免疫组织化学方法检测大鼠肿瘤组织中 ESM-1和 VEGF 的蛋白表达水平。结果与肿瘤对照组比较,小柴胡汤各剂量组大鼠血清及肿瘤组织中 ESM-1、VEGF 蛋白含量和蛋白表达水平均降低,蛋白含量随剂量增长降低趋势明显差异有统计学意义(P <0.01),蛋白表达水平 C组、D 组降低较为显著但二者之间差异无统计学意义(P >0.05);阳性药各剂量组大鼠血清及肿瘤组织中 ESM-1、VEGF 蛋白含量和蛋白水平亦均降低,且随剂量增长降低趋势明显差异亦有统计学意义(P <0.01);阳性药 A 组、小柴胡汤 C 组和 D 组这三组之间在肿瘤组织蛋白表达水平上差异无统计学意义(P >0.05)。结论与阳性药各剂量组的治疗机制相一致,小柴胡汤各剂量组均能通过下调 ESM-1和 VEGF 的水平治疗 C6胶质瘤,高剂量组作用最为显著,其作用机制有待于进一步探讨。

목적:탐토소시호탕대 C6효질류대서 ESM-1화 VEGF 단백표체수평적영향。방법청길급 Wistar 노년웅성대서18월령,n=60지,우전지액와피하접충2×106/20μl 적세포현액;연후수궤분위6조:종류대조조、양성약A 조(체막서알;50 mg/Kg)、양성약 B 조(체막서알;25 mg/Kg)、소시호탕 C 조(50 g/Kg)、소시호탕 D 조(20 g/Kg)、소시호탕 E 조(10 g/Kg);매조균분별관위급여생리염수、양성약 A、양성약 B、소시호탕 C、소시호탕 D 화소시호탕E,련속복약5 d,휴식4 d,공급약3차,우최후일차급약후2 d 처사동물,안구취혈급취종류조직。채용 ELISA 매련면역법검측대서혈청급종류조직중 ESM-1화 VEGF 적단백함량;채용면역조직화학방법검측대서종류조직중 ESM-1화 VEGF 적단백표체수평。결과여종류대조조비교,소시호탕각제량조대서혈청급종류조직중 ESM-1、VEGF 단백함량화단백표체수평균강저,단백함량수제량증장강저추세명현차이유통계학의의(P <0.01),단백표체수평 C조、D 조강저교위현저단이자지간차이무통계학의의(P >0.05);양성약각제량조대서혈청급종류조직중 ESM-1、VEGF 단백함량화단백수평역균강저,차수제량증장강저추세명현차이역유통계학의의(P <0.01);양성약 A 조、소시호탕 C 조화 D 조저삼조지간재종류조직단백표체수평상차이무통계학의의(P >0.05)。결론여양성약각제량조적치료궤제상일치,소시호탕각제량조균능통과하조 ESM-1화 VEGF 적수평치료 C6효질류,고제량조작용최위현저,기작용궤제유대우진일보탐토。
Objective To investigate Xiaochaihutang impact on ESM-1 and VEGF protein expression in C6 glioma in rats.Methods Wistar male rats aged 18 months,n=60,with the right forelimb axillary subcutaneous inoculation of cell suspension 2×106/20 μl;then randomly divided into six groups :control group tumors ,positive drug group A(temo-zolomide;50 mg/Kg),positive drug group B(temozolomide;50 mg/Kg),Xiaochaihutang group C(50 mg/Kg),Xi-aochaihutang group d(20 mg/Kg),Xiaochaihutang group E(10 mg/Kg);each group were orally administered saline, positive drug A,positive drug B,Xiaochaihutang C,Xiaochaihutang D and E,continuous medication 5 d,rest 4 d,were to drug three times in the last 2 d animals were sacrificed after administration ,blood and tumor tissue.Using ELISA to detect the protein content of ESM-1 and VEGF in serum and tumor tissue,Immunohistochemistry was used to detect the protein expression of ESM-1 and VEGF in rat tumor tissue.Results Compared with the control group tumors,the protein content and protein expression of ESM-1 and VEGF in Xiaochaihutang each dose group on serum and tumor tis-sue were decreased ,protein content reduced the growth trend with dose significant difference was statistically(P <0.01);protein expression levels group C,D group decreased more significant but the difference between them was not statistically sifnificant(P >0.05);positive serum and tumor tissue of rats in each dose group in protein content and protein levels of ESM-1 and VEGF were lower,and with lower doses of growth trend evident difference was also statis-tically significant(P <0.01);positive drug group A,Xiaochaihutang group C and D between the three groups ,the difference was not statistically significant(P >0.05).Conclusion Consistent with mechanism of positive drug treat-ment each dose group ,Xiaochaihutang each dose group could cut through the level of ESM-1 and VEGF treatment C6 glioma high-dose group the most significant effect,the mechanism needs further explored.
