目的:观察消痰和胃方对胃癌前病变大鼠胃黏膜组织中IL-1β蛋白及基因表达的影响。方法 Wistar大鼠50只随机分为正常组、模型组、验证组、维酶素组、中药组5组,每组10只。除正常组外其他4组采用高剂量N-甲基-N′硝基-N-亚硝基胍( MNNG)综合造模法干预32周建立大鼠胃癌前病变( PLGC)模型。验证组处死后取胃,验证造模成功,其余4组按照各自给药方法干预12周。Western Blot法检测4组大鼠胃黏膜组织中白细胞介素-1β( IL-1β)蛋白表达含量;RT-PCR法检测4组大鼠胃黏膜组织中IL-1β mRNA表达含量。结果模型组、维酶素组IL-1β蛋白及IL-1β mRNA表达较正常组显著升高( P﹤0.05,P﹤0.01)。中药组IL-1β蛋白表达较模型组、维酶素组下降( P﹤0.05),与正常组相比差异无统计学意义( P﹥0.05);中药组IL-1β mRNA平均相对表达量较模型组显著降低( P﹤0.01)。结论消痰和胃方可下调胃癌前病变大鼠IL-1β蛋白及基因表达。

목적:관찰소담화위방대위암전병변대서위점막조직중IL-1β단백급기인표체적영향。방법 Wistar대서50지수궤분위정상조、모형조、험증조、유매소조、중약조5조,매조10지。제정상조외기타4조채용고제량N-갑기-N′초기-N-아초기고( MNNG)종합조모법간예32주건립대서위암전병변( PLGC)모형。험증조처사후취위,험증조모성공,기여4조안조각자급약방법간예12주。Western Blot법검측4조대서위점막조직중백세포개소-1β( IL-1β)단백표체함량;RT-PCR법검측4조대서위점막조직중IL-1β mRNA표체함량。결과모형조、유매소조IL-1β단백급IL-1β mRNA표체교정상조현저승고( P﹤0.05,P﹤0.01)。중약조IL-1β단백표체교모형조、유매소조하강( P﹤0.05),여정상조상비차이무통계학의의( P﹥0.05);중약조IL-1β mRNA평균상대표체량교모형조현저강저( P﹤0.01)。결론소담화위방가하조위암전병변대서IL-1β단백급기인표체。
Objective To observe the effects of Xiaotan Hewei Formula on the expressions of inter-leukin-1β( IL-1β)and IL-1β mRNA in rats with precancerous lesions of gastric cancer( PLGC). Meth-ods Fifty Wistar rats were randomized into the normal group,model group,validation group,vitacoenzyme group and Chinese medicine group,with 10 in each. The N - methyl - N′- nitro - N - nitrosoguanidine ( MNNG)complex modeling method was used to establish the PLGC rat models in all groups except the nor-mal group. The rats in the validation group were sacrificed to verify whether the modeling was successful. The other four groups were intervened with the corresponding medication for 12 weeks. The expressions of IL-1βand IL-1β mRNA were measured. Results Comparing with the normal group,the expressions of IL-1βand IL-1βmRNA were significantly increased in the model group and vitacoenzyme group(P﹤0. 05 or P﹤0. 01). Comparing with the model group and vitacoenzyme group,the expression of IL-1β was significantly decreased in the Chinese medicine group(P﹤0. 05). There was no significant difference between the normal group and Chinese medicine group in the expression of IL-1β(P﹥0. 05). Comparing with the model group, the average relative expression of IL-1β mRNA was significantly decreased in the Chinese medicine group (P﹤0. 01). Conclusion Xiaotan Hewei Formula can down regulate the expressions of IL-1βand IL-1βmRNA in rats with PLGC.
