目的:研究益寿丸对D-半乳糖( D-gal)所致衰老小鼠线粒体损伤的影响,探讨其抗衰老的作用机制。方法采用 PCR法,考察益寿丸对D-半乳糖致衰老模型小鼠脑线粒体DNA缺失突变的影响;并在透射电镜下对衰老动物脑组织线粒体的超微结构进行观察。结果模型组、益寿丸低剂量组及阳性药组小鼠海马mtDNA中存在着约3.87kb片断缺失。益寿丸高、中剂量组未见缺失片断。结论提示益寿丸有防御模型小鼠脑mtDNA缺失突变的作用,益寿丸在调节能量代谢方面具有一定改善作用。
目的:研究益壽汍對D-半乳糖( D-gal)所緻衰老小鼠線粒體損傷的影響,探討其抗衰老的作用機製。方法採用 PCR法,攷察益壽汍對D-半乳糖緻衰老模型小鼠腦線粒體DNA缺失突變的影響;併在透射電鏡下對衰老動物腦組織線粒體的超微結構進行觀察。結果模型組、益壽汍低劑量組及暘性藥組小鼠海馬mtDNA中存在著約3.87kb片斷缺失。益壽汍高、中劑量組未見缺失片斷。結論提示益壽汍有防禦模型小鼠腦mtDNA缺失突變的作用,益壽汍在調節能量代謝方麵具有一定改善作用。
목적:연구익수환대D-반유당( D-gal)소치쇠로소서선립체손상적영향,탐토기항쇠로적작용궤제。방법채용 PCR법,고찰익수환대D-반유당치쇠로모형소서뇌선립체DNA결실돌변적영향;병재투사전경하대쇠로동물뇌조직선립체적초미결구진행관찰。결과모형조、익수환저제량조급양성약조소서해마mtDNA중존재착약3.87kb편단결실。익수환고、중제량조미견결실편단。결론제시익수환유방어모형소서뇌mtDNA결실돌변적작용,익수환재조절능량대사방면구유일정개선작용。
OBJECTIVE To study the YiShou Pills on brain mitochondrial DNA missing of the aging model mice ,to explore the mechanism of its anti-aging.METHODS PCR was used to inspect the effect of YiShou Pills on brain mitochondrial DNA missing of the aging model mice induced by D-gal;And ultrastructure of aging mice brain mitochondria were observed under the transmission electron microscope.RESLUTS The results showed that model group,YiShou Pills low dosage group and positive controlled group carried 3.87kb mtDNA deficiency in brain of mice.While YiShou Pills high and middle dosage group could not carried mtDNA deficiency.CONCLUSION YiShou Pills could defense the brain mitochondrial DNA missing of the aging model mice ,which had the improvement action of regulating energy metabolism.
