比较不同方法提取土鳖虫肠道菌群总 DNA 的差异,并对土鳖虫肠道菌群组成多样性进行分析。采用试剂盒法和 CTAB 法提取土鳖虫肠道菌群总 DNA,检测其浓度和纯度,扩增16 S DNA 进行鉴定,并利用 ERIC-PCR 技术对土鳖虫肠道菌群多样性进行比较分析。结果显示,两种方法均能提取 DNA,用于16 S DNA 的扩增和 ERIC-PCR 分析,可用于进行土鳖虫肠道菌群的分子生态研究,CTAB 法更能反应土鳖虫肠道菌群的多样性。

To screen a method that could provide a high quality DNA template and investigate the similarity and diversity of intestinal microflora in Eupolyphaga sinesis,TIANamp Stool DNA Kit method and CTAB method were used to extract total DNA from intestinal microflora of the Eupolyphaga sinesis.The DNA quality was compared based on the concentration,purity and 16 S DNA PCR products,and analysis of intestinal microflora was carried out by ERIC-PCR fingerprint.The results showed that total DNA ex-tracted by each of the two methods could be used to 16 S DNA amplification and ERIC-PCR fingerprint a-nalysis.The quality of total DNA extracted by the two methods was good enough to study molecular ecol-ogy of intestinal microflora.But the result of ERIC-PCR of DNA extracted by CTAB method showed more microflora diversities.
