为探索 Ca2＋与氧自由基对离体线粒体的损伤机制，采用不同浓度的 CaCl2和 H 2 O2诱导猪心线粒体损伤后，通过对线粒体内部超微结构的透射电子显微镜检测以及外部形态的原子力显微镜扫描来确定Ca2＋和氧自由基对离体猪心线粒体的损伤作用。结果可知，在一定浓度范围内，线粒体的损伤程度与 CaCl2和 H2 O2的损伤浓度呈正相关。不同浓度 Ca2＋和氧自由基可以不同程度的引起线粒体嵴分裂，线粒体膜的塌陷、破坏，基质的变化，差异明显。本试验通过透射电子显微镜和原子力显微镜对线粒体的观察可以得出线粒体被破坏过程中的各部分的超微结构变化的5阶段模型，为之后各类线粒体损伤保护试验提供依据。

The mitochondria were injured by different concentrations of CaCl2 and H 2 O2 to study the mecha-nism of the process of mitochondrial damage.The transmission electron microscope (TEM)and atomic force microscope (AFM)were used for observing the internal structure and scanning the external morphol-ogy respectively.The results demonstrated that different concentrations of Ca2+ and oxygen free radicals caused varying levels of mitochondrial swelling and collapse of mitochondrial membrane,and the difference is significant.In this study,we got many photos by the TEM and AFM,so we can establish a model of the process of 5 mitochondrial levels'damages to provide the foundation for the further researches on mito-chondria.
