目的采用凝胶色谱法测定大鼠口服麦冬多糖MDG-1后尿液及粪便中含量的变化。方法采用异硫氰酸荧光素(FITC)对麦冬多糖MDG-1进行标记。采用荧光凝胶色谱法(HPGPC)对SD-雄性大鼠口服MDG-1后的粪便及尿液内MDG-1含量进行测定。给药组按照150 mg/kg给予F-MDG-1,空白组给予纯水,分别在1、2、3、4、5、6、7 d后收集尿液及粪便,测定含量。结果在连续给予F-MDG-1量150 mg/kg后,尿液中含量测定在3 d时达到最大排泄量0.3133 mg;粪便中含量在2 d达到最大排泄量为5.70 mg。结论 MDG-1在肠道内被分解,并和肠道相互作用。采用FITC对MDG-1进行标记,并采用荧光色谱法对MDG-1在排泄物内的含量变化进行研究是可行的。

목적채용응효색보법측정대서구복맥동다당MDG-1후뇨액급분편중함량적변화。방법채용이류청산형광소(FITC)대맥동다당MDG-1진행표기。채용형광응효색보법(HPGPC)대SD-웅성대서구복MDG-1후적분편급뇨액내MDG-1함량진행측정。급약조안조150 mg/kg급여F-MDG-1,공백조급여순수,분별재1、2、3、4、5、6、7 d후수집뇨액급분편,측정함량。결과재련속급여F-MDG-1량150 mg/kg후,뇨액중함량측정재3 d시체도최대배설량0.3133 mg;분편중함량재2 d체도최대배설량위5.70 mg。결론 MDG-1재장도내피분해,병화장도상호작용。채용FITC대MDG-1진행표기,병채용형광색보법대MDG-1재배설물내적함량변화진행연구시가행적。
Objective To investigate the contents after oral Radix Ophiopogon japonicas polysaccharides MDG-1 in rats urine and faeces by HPGPC.Methods Labeling the MDG-1 and detecting replacement of FITC then using the HPGPC was in order to detect the contents of MDG-1 in urine and faeces after oral. The rats were given doses of F-MDG-1 150 mg.kg-1 then urine and faeces were harvested at predefined times (1,2,3, 4,5,6,7 d postdose;n=six per time point) then stored at-20℃until analysis. Results The F-MDG-1 in rats urine were described with the max 0.3133 mg in 3 d. The F-MDG-1 in rats faeces were described with the max 5.70 mg in 2 d.Conclusion In this study, F-MDG-1 was decomposition in the intestinal tract, F-MDG-1 and Gastrointestinal tract were affected each other. The combination of the FITC prelabeling method with the highperformance gel permeation chromatography should be helpful to other polysaccharides research in rats urine and faeces.
