目的:探讨人脐血源基质细胞(hUCBDSCs)对主要组织相容性复合体(MHC)半相合外周血造血干细胞移植小鼠移植物抗宿主病(GVHD)的影响及其可能机制。方法构建小鼠MHC半相合外周血造血干细胞移植GVHD模型,观察联合hUCBDSCs移植组和单纯移植组小鼠的生存率,临床表现,绘制Kaplan-Meier生存曲线;移植后14 d对小鼠皮肤、肝脏和小肠进行病理检测;流式细胞术分析移植后21 d小鼠脾脏中自然杀伤细胞(NK细胞)的比例。结果和对照组相比,实验组的GVHD临床表现明显减轻;Kaplan-Meier生存曲线显示实验组的生存率明显高于对照组;病理切片HE染色结果显示实验组的病理改变程度显著低于对照组;流式细胞术检测联合hUCBDSCs移植实验组NK细胞的阳性率为(26.34±2.7)%,对照组阳性率为(14.92±0.34)%,实验组显著高于对照组。结论 hUCBDSCs 可能通过提高 NK 细胞的比例减轻MHC半相合外周血造血干细胞移植小鼠GVHD临床表现和病理变化,提高小鼠生存率。

목적:탐토인제혈원기질세포(hUCBDSCs)대주요조직상용성복합체(MHC)반상합외주혈조혈간세포이식소서이식물항숙주병(GVHD)적영향급기가능궤제。방법구건소서MHC반상합외주혈조혈간세포이식GVHD모형,관찰연합hUCBDSCs이식조화단순이식조소서적생존솔,림상표현,회제Kaplan-Meier생존곡선;이식후14 d대소서피부、간장화소장진행병리검측;류식세포술분석이식후21 d소서비장중자연살상세포(NK세포)적비례。결과화대조조상비,실험조적GVHD림상표현명현감경;Kaplan-Meier생존곡선현시실험조적생존솔명현고우대조조;병리절편HE염색결과현시실험조적병리개변정도현저저우대조조;류식세포술검측연합hUCBDSCs이식실험조NK세포적양성솔위(26.34±2.7)%,대조조양성솔위(14.92±0.34)%,실험조현저고우대조조。결론 hUCBDSCs 가능통과제고 NK 세포적비례감경MHC반상합외주혈조혈간세포이식소서GVHD림상표현화병리변화,제고소서생존솔。
ObjectiveTo explore the effect of hUCBDSC on GVHD in mice after haploidentical peripheral blood hematopoietic stem cell transplantation and its mechanisms.Methods Peripheral blood hematopoietic stem cell and splenic cells were injected intravenously into each sublethally irradiated recipient mouse; Meantime, the clinical GVHD symptoms in mice cotransplanted hUCBDSC or not were observed;Kaplan- Meier survival curve was made; Pathological analysis was carried out on GVHD target tissues such as skin, liver and small intestine on day 14 post transplantation; The percentage of NK cells in spleen was determined by flow cytometry.ResultsIn group cotransplanted hUCBDSC, clinical GVHD symptoms were ameliorated and mortality was reduced compared to control group. Moreover, histopathological changes in experimental group were superior to control group. The percentage of splenic NK cells in experimental group was significantly higher than in control group on day21 post transplantation.ConclusionhUCBDSC may alleviate the incidence of GVHD in mice suffering from haploidentical peripheral blood hematopoietic cell transplantation by mechanisms of NK cells.
