目的：研究不同浓度胰岛素对高糖培养下视网膜色素上皮细胞增殖活力的影响。方法：体外培养人RPE(ARPE-19)，在高糖(30 mmol/L)浓度下给予0U/L，100U/L，101U/L，102U/L，103U/L，104U/L胰岛素作用12h、24h、48h后用MTT法检测各组视网膜色素上皮细胞的增生活力。结果：各浓度胰岛素组在不同观察时间（12小时、24小时、48小时），其吸光度值（OD570）与空白对照组相比，差异无统计学意义（P>0.05），各组吸光度值（OD570）随作用时间的延长而有增加的趋势，差异具有统计学意义（P<0.05）。结论：胰岛素对高糖培养下视网膜色素上皮细胞增殖活力无明显影响。

Objective:To investigate the effect of different concentrations of insulin on the proliferation of high glucose cultured human retinal pigment epithelial cells-19 in vitro. Methods: Human retinal pigment epithelial cells (ARPE-19) were cultured in vitro, exposed to high glucose group, treated with different concentrations of insulin (0U/L、100U/L、101U/L、102U/L、103U/L、104U/L) respectively. cellproliferation was detected by MTT assay after 12, 24 and 48 hours. Results: There was no statistical significant difference among different concentrations of insulin intervention conditions in groups of high glucose compared to controls (P> 0.05).There was a statistical y significant difference with the extension of time (P<0.05). Conclusion: Insulin had no effect on the proliferation of ARPE-19 cells in the high glucose condition.
