用响应面法优化繁缕[Stellaria media(L.) Cyr.]中总黄酮类化合物的提取工艺,通过羟自由基、超氧阴离子自由基、总还原力试验测定繁缕中总黄酮的体外抗氧化作用,并与抗坏血酸和叔丁基对苯二酚的抗氧化能力进行比较。结果表明,提取工艺最佳条件为提取温度86℃、提取时间2.5 h、液料比V 乙醇:m 繁缕粉为31.5:1(mL:g)、提取2次,该条件下总黄酮提取率预测值为3.907%,验证值为3.879%。繁缕中总黄酮的还原能力、对超氧阴离子自由基和羟自由基的清除能力均表现出较好的效果,清除超氧阴离子自由基和羟自由基的IC50分别为0.583 mg/mL和0.626 mg/mL,其抗氧化能力高于维生素C和叔丁基对苯二酚。

용향응면법우화번루[Stellaria media(L.) The conditions for extrating flavonoids from Stellaria media (L.) Cyr. were optimized by response surface methodology. The antioxidant activity in vitro was evaluated by hydroxyl radical scavenging assay, superoxide anion radical scavenging assay and total reducing power assay with vitamin C and tert-Butylhydroquinone as the reference. The results showed that the optimal extraction conditions were extraction temperature of 86℃, extraction time of 2.5 h, material-to-liquid ratio of 1:31.5 and extracted twice. Under the optimal condition, the predicted and actual values of flavonoids yield were 3.907% and 3.879%, respectively. The antioxidant test showed that Stellaria media (L.) Cyr. flavonoids had excellent reducing power and high abilities to scavenge superoxide anion and hydroxyl free radicals with IC50 values of 0.583 mg/mL and 0.626 mg/mL, respectively. The antioxidant effect of flavonoids from Stellaria media (L.) Cyr. were higher than that of VC and TBHQ.
