目的:研究通过建立纳米磁珠分选-时间分辨免疫荧光联合检测法(nanomagicbeabs sorting-time resolved fluoroimmunoassay,nano-TRFIA)对健康人血清半乳糖凝集素-3(Galectin-3)进行检测并与常规时间分辨免疫荧光法(time resolved fluoroimmunoassay,TRFIA)比较。方法:采用Galectin-3抗体标记的纳米磁珠对20例健康志愿者的外周血血清进行分选,并串联时间分辨免疫荧光进行检测。结果:使用nano-TRFIA检测出的Galectin-3血清质量浓度为(0.3803±0.3129) ng/mL,而使用常规TRFIA检测出的Galectin-3血清质量浓度为(0.2901±0.2783) ng/mL;nano-TRFIA检出值可达到皮克水平,与常规TRFIA比较,两者的检测灵敏度差异有统计学意义(P<0.05)。结论:nano-TRFIA够实现对血清标本的重复使用,并能提高对血清Galectin-3的检测灵敏度。

목적:연구통과건립납미자주분선-시간분변면역형광연합검측법(nanomagicbeabs sorting-time resolved fluoroimmunoassay,nano-TRFIA)대건강인혈청반유당응집소-3(Galectin-3)진행검측병여상규시간분변면역형광법(time resolved fluoroimmunoassay,TRFIA)비교。방법:채용Galectin-3항체표기적납미자주대20례건강지원자적외주혈혈청진행분선,병천련시간분변면역형광진행검측。결과:사용nano-TRFIA검측출적Galectin-3혈청질량농도위(0.3803±0.3129) ng/mL,이사용상규TRFIA검측출적Galectin-3혈청질량농도위(0.2901±0.2783) ng/mL;nano-TRFIA검출치가체도피극수평,여상규TRFIA비교,량자적검측령민도차이유통계학의의(P<0.05)。결론:nano-TRFIA구실현대혈청표본적중복사용,병능제고대혈청Galectin-3적검측령민도。
Objecti ve: To establish nanomagicbeabs sorting-TRFIA tandem analysis method for the detection of serum Galectin-3 for healthy people and comparison with the conventional time resolved fluoroimmunoassay method. Methods:Serum levels of Galectin-3 were measured by application of nanomagicbeabs sorting with monoclonal anti-human Galectin-3 labeled antibody and tandem TRFIA in healthy volunteers. Results:Serum levels of Galectin-3 in healthy volunteers by nano-TRFIA was (0.380 3±0.312 9) ng/mL, and that by conventional TRFIA was (0.290 1±0.278 3) ng/mL. The value of Galectin-3 by nano-TRFIA can reach picogram level. There was a significant difference between nano-TRFIA and conventional TRFIA on detection sensitivity (P<0.05). Conclusion: Nanomagicbeabs sorting-TRFIA tandem analysis method has the ability to use repeatly of serum specimens and to improve the detection sensitivity of serum Galectin-3.
