利用中蜂囊状幼虫病毒(CSBV)灭活疫苗作为抗原制备兔多克隆抗体。利用RT- PCR方法对疑似感染CSBV的中蜂样品进行检测,检测阳性后,经差速离心法纯化CSBV,并用BCA方法检测病毒浓度,将病毒浓度稀释至0.25 mg/mL。按稀释后的CSBV体积的0.3%加入甲醛灭活进行灭活,制备CSBV油乳剂灭活苗,经无菌、安全性和稳定性检验合格后,对新西兰大白兔进行3次免疫,每次间隔2周,对其产生的多克隆抗体进行了SDS- PAGE分析和效价检测,结果表明制备的兔多克隆抗体效价OD值最高可达到1.109。实现了中蜂囊状幼虫病毒高效价多克隆抗体的制备,为深入研究CSBV和综合防治奠定了基础。

To develop a safe, effective and quality-controllable antibody against Chinese sac-brood virus for preventing and treating Chinese sacbrood virus. Suspected been larvaes were identi-fied by RT-PCR technology, and positive samples were purified by CSBV, and virus concentration were detected by BCA, then diluted to the concentration of 0.25 mg/ml. Chinese sacbrood virus are inactivated by 0.3% formaldehyde, and oil emulsion inactivated vaccine were produced after sterili-ty, safety, and stability determination. Then the New Zealand White Rabbit were injected with the vaccine for 3 times, once two weeks in order to analysis the polyclonal antibody against Chinese sacbrood virus. The results showed that the OD of the antibody titer is as high as 1.109. The high titer vaccine is prepared,and the research provide foundation for the prevention of CSBV.
