目的:分离纯化刺糖多糖,并对刺糖多糖的结构进行基础性分析。方法采用水提醇沉法提取刺糖多糖,联合使用大孔吸附树脂、纤维素和葡聚糖凝胶柱色谱对刺糖多糖进行分离纯化,采用凝胶过滤法测定其纯度和相对分子量,气相色谱法分析其单糖组成,对含量较高的刺糖多糖组分进行部分酸水解、高碘酸氧化和Smith降解分析。结果经分离纯化得到11个精制多糖组分,相对分子量为2~10 kDa ,单糖组成多为鼠李糖、阿拉伯糖、甘露糖、葡萄糖和半乳糖。A P1-3的主链单糖残基为鼠李糖、甘露糖和半乳糖,末端残基和分支单糖残基为葡萄糖;A P2-3的主链单糖残基为鼠李糖、阿拉伯糖、葡萄糖和半乳糖,末端残基和分支单糖残基为鼠李糖、阿拉伯糖和甘露糖。结论采取的分离纯化方法切实可行。经结构分析可知刺糖多糖各组分多为有分枝的杂多糖。
目的:分離純化刺糖多糖,併對刺糖多糖的結構進行基礎性分析。方法採用水提醇沉法提取刺糖多糖,聯閤使用大孔吸附樹脂、纖維素和葡聚糖凝膠柱色譜對刺糖多糖進行分離純化,採用凝膠過濾法測定其純度和相對分子量,氣相色譜法分析其單糖組成,對含量較高的刺糖多糖組分進行部分痠水解、高碘痠氧化和Smith降解分析。結果經分離純化得到11箇精製多糖組分,相對分子量為2~10 kDa ,單糖組成多為鼠李糖、阿拉伯糖、甘露糖、葡萄糖和半乳糖。A P1-3的主鏈單糖殘基為鼠李糖、甘露糖和半乳糖,末耑殘基和分支單糖殘基為葡萄糖;A P2-3的主鏈單糖殘基為鼠李糖、阿拉伯糖、葡萄糖和半乳糖,末耑殘基和分支單糖殘基為鼠李糖、阿拉伯糖和甘露糖。結論採取的分離純化方法切實可行。經結構分析可知刺糖多糖各組分多為有分枝的雜多糖。
목적:분리순화자당다당,병대자당다당적결구진행기출성분석。방법채용수제순침법제취자당다당,연합사용대공흡부수지、섬유소화포취당응효주색보대자당다당진행분리순화,채용응효과려법측정기순도화상대분자량,기상색보법분석기단당조성,대함량교고적자당다당조분진행부분산수해、고전산양화화Smith강해분석。결과경분리순화득도11개정제다당조분,상대분자량위2~10 kDa ,단당조성다위서리당、아랍백당、감로당、포도당화반유당。A P1-3적주련단당잔기위서리당、감로당화반유당,말단잔기화분지단당잔기위포도당;A P2-3적주련단당잔기위서리당、아랍백당、포도당화반유당,말단잔기화분지단당잔기위서리당、아랍백당화감로당。결론채취적분리순화방법절실가행。경결구분석가지자당다당각조분다위유분지적잡다당。
Objective To isolate ,purify the polysaccharide from Alhagi-honey and clarify its basic chemical structure .Methods:Method of water extracting-alcohol precipitating was applied to extract polysaccharide from Alhagi-honey .Macroporous adsorption resin chromatography ,DEAE cellulose chromatography and Sephadex gel chromatography were applied to fractionalize and purify the polysaccharide .The homogeneity and molecular weight of the polysaccharides were determined by Gel filtration .The monosaccharide com-positions of the polysaccharides were identified by Gas chromatography (GC ) . Partial hydrolysis with acid ,periodate oxidation and smith degradation were used to analyze the chemical structure of polysaccha-ride fractions which is of high content in alhagi-honey .Results 11 Polysaccharide fractions were gotten from Alhagi-honey . The results show that they were homogeneous polysaccharides and the molecular weights were between 2 kDa and 100 kDa .They were composed of rhamnose ,arabinose ,mannose ,glu-cose and galactose .The main chain of AP1-3 is mainly made up of rhamnose ,mannose ,and galactose . The side chain is composed of glucose ;The main chain of AP2-3 is mainly made up of rhamnose ,arabi-nose ,glucose and galactose .The side chain is composed of rhamnose ,arabinose and mannose .Conclusion <br> It have been proved that the eleven polysaccharide fractions were pure and homogeneous ,so the methods we have used to isolate and purify polysaeeharides were feasible and workable .Structural elucidation show that most of Alhagi-honey polysaccharide fractions were heteropolysaccharide .
