为研制安全有效的适合预防吉林地区PRRS的亚单位疫苗,利用吉林地区PRRS病猪肺脏中分离获得的PRRSVJL-0605病毒株,参照GeneBank中已发表的PRRSV序列设计1对特异性引物,克隆M蛋白基因,根据M蛋白基因序列和推导的氨基酸序列进行同源性、系统进化树、疏水域及抗原表位的分析。结果表明,PRRSVJL-0605株的M蛋白氨基酸序列与CH—la和HEBl株高度一致,而与VR-2332和BJ-4株有一定的差异,和LV株在N端及中心区具有很大的差异,JL-0605株M蛋白抗原性及亲水性与中国河北株具有较高的相似性;Jb0605株与V~2332株相比在60位处抗原表位优势减弱。本研究为PRRSM蛋白和基因的深入研究及开发利用提供科学依据。

In order to study and find appropriate PRRSV subunit vaccine for native place, used specific primers which were designed according to the PRRSV sequence published by Genebank, and amplified and cloned the DNA of major structural proteins M of isolate plant JL-0605 separated from PRRS diseased pig lungs in Jilin Province. Then amino acid sequence was analyzed in homology, phylogenetic tree and epitope which was deduced by the separated strain protein M. The results showed that M protein amino acid se- quence of plant PRRSV JL-0605 has the same height with plant CH-la and plant HEB1, but a little differ- ent with plant VR-2332 and plant BJ-4, especially with plant LV in the protein N and middle. The antigen- icity and hydrophile of protein M, plant JL-0605, are much similar with the plant from Hebei province, China. But compared with plant VR-2332, the superiority of plants JL-0605 is weakened in amino acids sequence 60. The above research laid the foundation for analysis of PRRS M protein specific immune, and provide an important means for the screening of effective antigen and development of prevention preparation.
