为探索贯叶连翘愈伤组织和不定根反应器大量扩繁的最佳条件,以贯叶连翘种子为材料,对愈伤组织的诱导和增殖条件进行优化,并对不同培养条件下愈伤组织和不定根增殖生长情况进行调查。结果表明,愈伤组织在附加吲哚丁酸(IBA)4mg/L,激动素(KT)0.02mg/L和蔗糖30g/L的MS培养基中增殖效果佳。无论是愈伤组织还是不定根的增殖,反应器培养效果好于固体培养和振荡培养。在生物反应器内培养不定根的过程中,研究了培养基中pH值对不定根增殖生长和不定根中金丝桃素含量的影响,结果pH值为4.8~7.8时不定根生长不受pH值的影响,但金丝桃索的含量在pH值为4.8和5.8处理中显著高于pH值为6.8和7.8的处理,金丝桃素生产量也在pH值为4.8和5.8处理中较高(7.09和6.77mg/L)。说明利用反应器进行愈伤组织和不定根培养是短期内获得有效物质的有效方法之一。

To explore the optimum conditions for large scale propagation of callus and adventitious roots of Hypericum perforatum. The seeds of Hypericurn perforatum was used as the experimental material to op- timize the conditions for induction and propagation of callus, and the propagation growth of callus and ad- ventitious roots with different culture conditions was investigated. The results showed that callus propa- gatd best with IBA 4 mg/L, KT 0.02 mg/L and sucrose 30 g/L in MS medium. Whether callus or adven- titious roots propagation, bioreactor culture was better than solid and shaking culture. During bioreactor culture of adventitious roots of Hypericum perforaturn, the effect of medium pH on the propagation and hypericin content of adventitious roots were also investigated. Adventitious roots growth was not affected by the pH value from 4.8 to 7.8. Hypericin content in the treament of pH 4.8 and 5.8 were significantly higher than that in pH 6.8 and 7.8. Hypericin production was also higher in the treament of pH 4.8 and 5.8 (7.09 and 6.77 mg/L). It indicates that bioreactor culture of callus and adventitious roots is one of the effective methods for producting active substances in the short term.
