采用生物化学方法中的有机溶剂分级沉淀法,对碱性磷酸酶进行了分离纯化,用金氏法测定其活性,并用Folin-酚法测定酶含量,然后筛选碱性磷酸酶的最适pH值和最适温度。在此条件下研究不带电荷极性氨基酸对兔肝中碱性磷酸酶活性的影响。结果表明,兔肝中碱性磷酸酶的最适pH值为9.84,最适温度为39℃。在不带电荷极性氨基酸中甘氨酸、苏氨酸、半胱氨酸和天冬酰胺对碱性磷酸酶的活性有不同的抑制作用,而丝氨酸对碱性磷酸酶的活性有激活作用,这说明不带电荷极性氨基酸对酶活性抑制或激活作用与其侧链基团有关。
採用生物化學方法中的有機溶劑分級沉澱法,對堿性燐痠酶進行瞭分離純化,用金氏法測定其活性,併用Folin-酚法測定酶含量,然後篩選堿性燐痠酶的最適pH值和最適溫度。在此條件下研究不帶電荷極性氨基痠對兔肝中堿性燐痠酶活性的影響。結果錶明,兔肝中堿性燐痠酶的最適pH值為9.84,最適溫度為39℃。在不帶電荷極性氨基痠中甘氨痠、囌氨痠、半胱氨痠和天鼕酰胺對堿性燐痠酶的活性有不同的抑製作用,而絲氨痠對堿性燐痠酶的活性有激活作用,這說明不帶電荷極性氨基痠對酶活性抑製或激活作用與其側鏈基糰有關。

Alkaline phosphatase (AP) was separated and depurated with fractional precipitation of organic sol- vent from liver of rabbit. The AP activity was measured with the King' s Method, and the content of enzyme was detected with the Folin-phenol' s Method, in order to determine the optimum pH and temperature, for AP activity determination and investigate the effects of polar uncharged amino acids on liver AP activity of rabbit. The results showed that the optimum pH and temperature for AP activity determination was 9.84 and 39℃ , respectively. Threonine, cysteine, aspartic acid, and glutanic acid had inhibitory effects on liver AP activety, and serine had activated effect on liver AP activity. These results impiied that the actions of polar uncharged a- mino acids on liver AP activity were related with their side chains.
