侵染后抗性胚状体诱导频率低是影响橡胶树遗传转化发展的主要原因.而橡胶树次生体胚发生体系具有体胚增殖效率及植株再生率高的优点。以3个综合性状优良的橡胶树无性系次生体细胞胚状体为受体,研究其对根癌农杆菌的耐侵染能力、GU$基因的瞬时表达情况、探讨不同无性系胚状体对农杆菌转化的反应。结果表明:3个无性系农杆菌污染情况无显著差异,长菌的外植体经无菌水和抗生索水洗涤3d后即可恢复生长;侵染后每30个胚褐化了1.2~5个胚.占侵染总胚数的4.00%16.67%,品种间褐化面积无显著差异;热研87—6—62的总蓝斑数显著高于其它2个无性系.平均每个胚染上14.29个蓝斑,热研7—33—97和PRl07稍差,但每个胚也能分别染上8.14和8.72个蓝斑。因此3个无性系均可以胚状体作为遗传转化的受体。此结果对利用胚状体为受体改良橡胶树优良无性系奠定良好基础。
侵染後抗性胚狀體誘導頻率低是影響橡膠樹遺傳轉化髮展的主要原因.而橡膠樹次生體胚髮生體繫具有體胚增殖效率及植株再生率高的優點。以3箇綜閤性狀優良的橡膠樹無性繫次生體細胞胚狀體為受體,研究其對根癌農桿菌的耐侵染能力、GU$基因的瞬時錶達情況、探討不同無性繫胚狀體對農桿菌轉化的反應。結果錶明:3箇無性繫農桿菌汙染情況無顯著差異,長菌的外植體經無菌水和抗生索水洗滌3d後即可恢複生長;侵染後每30箇胚褐化瞭1.2~5箇胚.佔侵染總胚數的4.00%16.67%,品種間褐化麵積無顯著差異;熱研87—6—62的總藍斑數顯著高于其它2箇無性繫.平均每箇胚染上14.29箇藍斑,熱研7—33—97和PRl07稍差,但每箇胚也能分彆染上8.14和8.72箇藍斑。因此3箇無性繫均可以胚狀體作為遺傳轉化的受體。此結果對利用胚狀體為受體改良橡膠樹優良無性繫奠定良好基礎。

Low resistant embryo induction frequency after infection is the main reason affects the development of genetic transformation of Hevea, secondary somatic embryogenesis with high frequency in embryo multiplication and plant regeneration. In this study, secondary somatic embryos from three elite clones were used as receptor materials to study their reaction on Agrobacteriurn infection, including Agrobacterium contamination, browning and GUS transient expression. Results showed that there was no significant difference on Agrobacterium contamination and browning area between three clones, contaminated explants can restore after 3 days washing by sterile water followed by antibiotic water, and browning frequency was 4.00-16.67%, that is say 1.2-5 embryos browned after 30 embryos infected. The total number of CATAS 87-6-62 blue spots was significantly more than the other two clones, CATAS 7-33-97 and PR 107 slightly worse, but each embryo could also stained with 8.14 and 8.72 bh/e spots respectively. Therefore the three clones can use somatic embryos as a recipient for genetic transformation in Hevea. Results laid a good foundation for rubber tree elite clones improvement by using embryo as receptor.
