以反相毛细管液相色谱柱为例设计了一组简便易行的液相色谱柱填充效果评价实验。选用方便易溶的常用试剂丙酮作为死时间标记物兼作探针化合物,甲醇作为流动相,对毛细管C18柱进行了塔板高度一流速关系曲线的测定,取得了良好的效果。该色谱操作条件具有普适性,测试方便,适合于色谱实验教学和色谱工作者的填充柱效测试。

Abstract-A user-friendly experiment for evaluating column packing efficiency was designed, and it was also generally applicable to traditional reversed-phase HPLC packed column efficiency evaluation. Acetone was used as the dead time marker and methanol was used as mobile phase, then good results were achieved in the plate height - mobile phase velocity curve test. The chromatographic operating conditions are universal, and this experiment is suitable for HPLC teaching and the practical application of chromatography.
