以蓝狐( Alopex lagopus)胆囊胆汁为实验材料,采用正交试验建立了测定狐胆汁中结合型胆汁酸成分的高效液相色谱分析法。结果表明,采用Hypersil BDS C18(4.6 mm×250 mm,5μm)为色谱柱,V(甲醇)∶V(30 mmol/L磷酸二氢钾)=70∶30的溶液(pH=3.8)为流动相进行等度洗脱,流速为1.0 mL/min,检测波长为205 nm,柱温为25℃。测得狐胆汁中含有牛磺胆酸和牛磺鹅去氧胆酸,它们的峰面积比分别为35.05%和9.23%,不含牛磺熊去氧胆酸。各组分在3.33~10 g/L范围内具有良好的线性关系。本方法具有简单快速、分离效果好、重复性好等优点,可以用来分析狐胆汁中结合型胆汁酸。

An experiment was conducted to build the method for compositing conjugated bile acid in fox bile by high performance liquid chromatography through orthogonal test with blue fox ( Alopex lagopus) gallbladder bile as experimental material. The column was Hypers-il BDS C18(4.6 mm×250 mm,5 μm).The solution of methanol/30 mmol/L potassium dihydrogen phosphate (70 ∶30, PH=3.8) was used as mobile phase in isocratic elution.Flow rate was 1.0 mL/min.The detector wavelength was 205 nm and the column temperature was 25℃.The fox bile contained taurocholic acid and taurochenode-oxycholic acid and their peak areas are 35.05%and 9.23%, respectively, without tauroursodeoxycholic acid.Each com-ponent has a good liner relationship in 3.33-10 g/L.The method is simple and rapid with accurate separation and good re-producibility.It can be used for analyzing conjugated bile acid in fox bile.
