马铃薯再生体系的建立是进行马铃薯遗传转化的前提条件。本研究以马铃薯克新1号为植物材料,对马铃薯愈伤组织的诱导、不定芽的分化进行了最佳培养基优化。结果表明,马铃薯诱导愈伤组织的最佳培养基激素浓度是2.5mg/L6-BA、0.2mg/LNAA,且茎段的诱导率要高于叶片的诱导率。不定芽分化的最佳培养基激素浓度为2.0mg/L6-BA、0.1mg/LNAA、5.0mg/LGA3,诱导率为30%,诱导的不定芽在0.2mg/LN从生根培养基中可以正常生根。本研究初步建立了马铃薯再生体系,为后续的金黄色葡萄球菌奶牛乳腺炎转基因马铃薯疫苗的制备奠定了基础。

마령서재생체계적건립시진행마령서유전전화적전제조건。본연구이마령서극신1호위식물재료,대마령서유상조직적유도、불정아적분화진행료최가배양기우화。결과표명,마령서유도유상조직적최가배양기격소농도시2.5mg/L6-BA、0.2mg/LNAA,차경단적유도솔요고우협편적유도솔。불정아분화적최가배양기격소농도위2.0mg/L6-BA、0.1mg/LNAA、5.0mg/LGA3,유도솔위30%,유도적불정아재0.2mg/LN종생근배양기중가이정상생근。본연구초보건립료마령서재생체계,위후속적금황색포도구균내우유선염전기인마령서역묘적제비전정료기출。
Establishment of regeneration system is a prerequisite of genetic transformation of potato. In this study, the mediums for callus induction and shoot regeneration of potato were optimized with potato Kexin 1 as material. The results showed that the best hormone concentration for callus induction of potato was 2.5rag / L 6 - BA and O. 2 mg/ L NAA, and the induction rate of the stern section was higher than the leaves. The best hormone concentration for shoot regeneration of potato was 2.0rag / L 6 - BA, 0. ling / L NAA, and 5.0 mg / L of GA3, and the induction rate was about 30%, and furthermore, the shoots could root normally in the 0.2 mg / L NAA MS medium. In this study, the regeneration system of potato was established preliminarily, which laid the foundation for the preparation of transgenic potato vaccine against Staphylococcus aureus mastitis.
