目的:鉴定游离脂肪酸(free fatty acids ,FFA)对肝星状细胞(HSC)凋亡的影响。方法体外培养活化人HSC LX-2,加入FFA处理后,TRAIL诱导凋亡,通过流式细胞术检测细胞凋亡情况。通过caspase-3活性检测试剂盒检测细胞内caspase-3的活化情况,通过Q-PCR检测细胞内凋亡相关基因的表达情况。结果 FFA 能够抑制 TRAIL诱导的活化LX-2细胞凋亡,降低细胞内caspase-3的活性,Q-PCR的分析表明,FFA 能够影响细胞内凋亡相关基因BAX的表达,下调死亡受体TRAIL-R2和FAS的表达。结论体外FFA处理,能够抑制活化 HSC的凋亡。

목적:감정유리지방산(free fatty acids ,FFA)대간성상세포(HSC)조망적영향。방법체외배양활화인HSC LX-2,가입FFA처리후,TRAIL유도조망,통과류식세포술검측세포조망정황。통과caspase-3활성검측시제합검측세포내caspase-3적활화정황,통과Q-PCR검측세포내조망상관기인적표체정황。결과 FFA 능구억제 TRAIL유도적활화LX-2세포조망,강저세포내caspase-3적활성,Q-PCR적분석표명,FFA 능구영향세포내조망상관기인BAX적표체,하조사망수체TRAIL-R2화FAS적표체。결론체외FFA처리,능구억제활화 HSC적조망。
Objective To explore the effect of free fatty acids (FFAs) on the apoptosis of cultured hepatic stellate cells .Methods Activated human hepatic stellate cell (LX-2) line was induced apoptosis by TRAIL after being treated with FFAs .Apoptosis was detected by FACS .Intracellular caspase-3 activity was determined by caspase-3 assay kit . The expression of cell apoptosis-related genes was analyzed by real time-PCR .Results FFAs inhibited the apoptosis of activated LX-2 cell induced by TRAIL .Cellular activity of caspase3 was decreased by FFAs treatment .Q-PCR analysis showed that FFAs could decrease the expression of BAX , FAS and TRAIL-R2 . Conclusion FFAs could inhibit apoptosis of activated hepatic stellate cells in v itro .
