为探讨马立克氏病病毒强、弱毒株致癌基因Meq在感染过程中的变化规律,本研究用超强毒株RB1B与疫苗株CVI988分别感染次代鸡胚成纤维细胞(chicken embryo fiber cell,CEF),用荧光定量PCR方法检测Meq的变化规律。结果发现,疫苗株CVI988的Meq在感染细胞内持续上调,而超强毒株RB1B的Meq在感染细胞内72 h前为逐渐上调,然后下调;动物感染试验结果发现,超强毒株RB1B的Meq在鸡胸腺和脾脏组织内均表现为感染d21出现一个表达高峰,而疫苗株CVI988的Meq在SPF鸡胸腺中d14出现高峰,然后下降。有趣的是CVI988的Meq在脾脏组织中d14表达最低,然后持续升高。本研究为进一步研究Meq在马立克氏病病毒感染过程中的致瘤机理提供重要资料。

To illustrate dynamics of Meq gene expression during Marek's disease virus inefection in vivo and in vitro, chicken embryo fiber (CEF) cells and chickens were infected with very virulent strain RB1B or vaccine strain CVI988 and Meq levels were measured in real-time PCR. In CEF cells infected with vaccine strain CVI988, Meq expression continuously increased within 24 hours post infection (hpi). However, Meq gene expression increased from 24 hpi to 72 hpi and then decreased in CEF cells infected with very virulent strain RB1B. The results of chicken experiment showed that Meq expression in thymus and spleen gradually increased and peaked at day 21 post infection with very virulent strain RB1B. The Meq gene expression showed a different pattern in chickens inoculated with vaccine strain CVI988. In these chickens, Meq levels peaked in thymuses at day 14 post inoculation but were the lowest in spleens at the same time and gradually rose. These findings would contribute to better understand the dynamics of Meq gene expression during Marek's disease.
