以红薯叶为原料,选用D101、AB-8、DM130大孔吸附树脂进行静态吸附及解吸试验,筛选出适合吸附分离红薯叶总黄酮的树脂并确立其最佳吸附及解吸工艺参数。试验结果表明,AB-8大孔吸附树脂对红薯叶总黄酮吸附、解吸效果较好,最佳吸附条件为上样液浓度0.3 mg/mL,上样液流速2 mL/min,上样液pH 5.0,吸附率为66.8%;最佳解吸条件为洗脱液浓度90%,洗脱液流速4 mL/min,洗脱液用量5 BV,解吸率为70.3%。

이홍서협위원료,선용D101、AB-8、DM130대공흡부수지진행정태흡부급해흡시험,사선출괄합흡부분리홍서협총황동적수지병학립기최가흡부급해흡공예삼수。시험결과표명,AB-8대공흡부수지대홍서협총황동흡부、해흡효과교호,최가흡부조건위상양액농도0.3 mg/mL,상양액류속2 mL/min,상양액pH 5.0,흡부솔위66.8%;최가해흡조건위세탈액농도90%,세탈액류속4 mL/min,세탈액용량5 BV,해흡솔위70.3%。
Static adsorption and desorption capacity were assayed to screen the proper macroporous resin for total flavonoids from sweet potato leaves, and most suitable manipulation parameters of macroporous resin were confirmed. The results showed that AB-8 macroporous resin had strong adsorption and desorption capacity among the three resins used in this experiment. The most appropriate adsorption conditions were:sample concentration of 0.3 mg/mL, sample flow velocity of 2 mL/min, pH 5.0, and the adsorption rate reached 66.8%. The most fitting elution conditions were:ethanol concentration of 90%, elution rate of 4 mL/min, elution volume 5 BV, and the desorption rate was 70.3%.
