目的:探讨环氧化物酶-2(Cox-2)和肝细胞核因子-3β(HNF-3β)在非小细胞肺癌中的表达及其临床意义。方法采用免疫组化法检测 Cox-2和 HNF-3β在60例非小细胞肺癌和50例癌旁正常肺组织中的表达情况。结果60例非小细胞肺癌中 Cox-2和 HNF-3β阳性率分别为78.33%(47/60)和18.00%(9/60),50例癌旁正常肺组织 Cox-2和 HNF-3β阳性率分别为35.00%(21/50)和84.00%(42/50)。Cox-2在非小细胞肺癌与癌旁正常肺组织中的阳性率比较,前者明显高于后者,HNF-3β的阳性表达率反而明显低于癌旁正常肺组织,Cox-2和 HNF-3β蛋白表达呈负相关(P <0.05);Cox-2高表达和 HNF-3β低表达与非小细胞肺癌 TNM 临床分期、病理类型、有无淋巴结转移及患者生存期呈显著相关(P <0.05)。结论Cox-2和 HNF-3β在非小细胞肺癌的发生、发展过程中起了重要作用,推测 Cox-2和 HNF-3β蛋白表达可共同作为非小细胞肺癌早期诊断、预后评估的生物学标志物。为进一步研究非小细胞肺癌的发生发展机制,探寻新的靶向治疗途径提出指导意义。

Objective To observe the expressions and to discuss the clinical significances of cyclooxygen-ase-2 (COX-2)and hepatocyte nuclear factor-3β(HNF-3β)in non-small cell lung cancer (NSCLC). Methods An immunohistochemical staining was used to assay expressions of HNF-3βand COX-2 in 60 ca-ses of NSCLC and 50 cases of tumor-adjacent normal lung tissues. Results Out of 60 NSCLC cases,47 (78.33%)showed the positive expression of COX-2 and 9 (18.00%)showed the positive expression of HNF-3β.Out of 50 tumor- adjacent normal lung tissues,21(35.00%)showed the expression of COX-2 and 42 (84.00%)showed the expression of HNF-3β.The expression of Cox-2 was higher in NSCLC than those in tumor-adjacent normal lung tissues.The expression of HNF-3βwas lower in NSCLC than those in tumor-adjacent normal lung tissues.Cox-2 had negative association with the expression of HNF-3β protein (P <0.05).The high expression of Cox-2,low expression of HNF-3βin non-small cell lung cancer was positive-ly correlated with TNM clinical stage,pathological types,lymph node metastasis and survival (P <0.05). Conclusion COX-2 and HNF-3βplay very important roles in the occurrence and progress of NSCLC.COX-2 and HNF-3βmay become biomarkers of early diagnosis and prognostic evaluation for NSCLC,which pro-vides experimental data for further study of NSCLC occurrence and progress and for exploring a new target therapy of NSCLC.
