目的:探究化痰散结方对乳腺癌MCF-7/ADM移植瘤的逆转耐药机制。方法制备荷瘤小鼠模型;设立生理盐水空白对照组、阿霉素组、阿霉素+维拉帕米组、阿霉素+中药低中高剂量组,观察肿瘤组织内阿霉素蓄积情况,用流式细胞仪( FCM )检测各组移植瘤耐药细胞P糖蛋白( P-gp)表达情况。结果中药组及维拉帕米组均能减轻瘤重,提高抑瘤率,与生理盐水组及阿霉素组比较均有统计学意义(P<0.05),中药组及维拉帕米组均能够降低P-gp含量、提高细胞内阿霉素浓度,其中高剂量中药组对细胞内阿霉素、P-gp的影响较维拉帕米组差异有显著性( P<0.05)。结论化痰散结方能逆转阿霉素耐药,增减肿瘤组织中阿霉素含量,并呈剂量依赖性,同时降低P-gp的表达。

목적:탐구화담산결방대유선암MCF-7/ADM이식류적역전내약궤제。방법제비하류소서모형;설립생리염수공백대조조、아매소조、아매소+유랍파미조、아매소+중약저중고제량조,관찰종류조직내아매소축적정황,용류식세포의( FCM )검측각조이식류내약세포P당단백( P-gp)표체정황。결과중약조급유랍파미조균능감경류중,제고억류솔,여생리염수조급아매소조비교균유통계학의의(P<0.05),중약조급유랍파미조균능구강저P-gp함량、제고세포내아매소농도,기중고제량중약조대세포내아매소、P-gp적영향교유랍파미조차이유현저성( P<0.05)。결론화담산결방능역전아매소내약,증감종류조직중아매소함량,병정제량의뢰성,동시강저P-gp적표체。
Objective To explore the Compound of Reducing Phlegm and Resolving Masses on MCF-7/ADM breast cancer re-versal of drug resistance mechanism .Methods Preparation of tumor bearing mice model .Set up the normal control group ,driamycin group , doxorubicin+verapamil group ,driamycin+internal medicine low and high dose group were observed in tumor tissue accumulation of doxoru -bicin case,by flow cytometry(FCM) to detect the drug-resistant tumor cells P-glycoprotein(P-gp) expression.Results The traditional Chi-nese medicine group and the verapamil group reduced the tumor weight ,improve the tumor inhibition rate ,compared with the normal saline group and doxorubicin group were statistically significant (P<0.05),Chinese medicine group and the verapamil group were able to reduce the content of P-gp,increase the intracellular doxorubicin concentration ,the high dose group in reducing effect on doxorubicin ,P-gp cells the verapamil group with significant difference (P<0.05).Conclusion The Compound of Reducing Phlegm and Resolving Masses can reverse the resistance to doxorubicin concentration of adriamycin ,increase or decrease in tumor tissues ,and there are dose dependent ,and reducing the expression of P-gp.
