目的:探讨银杏叶提取物是否可以促进大鼠局灶性脑缺血后梗死周围区的血管新生,寻求促使脑缺血后血管新生、开发脑储备能力的有潜力的中医药疗法。方法采用线栓法建立大鼠大脑中动脉闭塞(MCAO)模型,术后24h采用银杏叶注射液以10mg/(kg体重·d)的剂量连续腹腔注射治疗6d,于缺血后第7 d、14天分别观察银杏叶组和对照组大鼠脑梗死体积、脑梗死灶周围血管密度(层粘连蛋白,Laminin的表达)和神经功能缺失评分的变化。结果与对照组比较,大鼠局灶性脑缺血后第7天和14天银杏叶组的梗死体积明显减小,梗死周围血管密度较对照组明显增加,并显示出更好的神经功能评分(P<0.05)。结论银杏叶提取物可促进局灶性脑缺血后大鼠脑梗死周围区的血管新生,从而开发脑储备能力,这可能是银杏叶提取物改善脑缺血后神经功能恢复的机制之一。
目的:探討銀杏葉提取物是否可以促進大鼠跼竈性腦缺血後梗死週圍區的血管新生,尋求促使腦缺血後血管新生、開髮腦儲備能力的有潛力的中醫藥療法。方法採用線栓法建立大鼠大腦中動脈閉塞(MCAO)模型,術後24h採用銀杏葉註射液以10mg/(kg體重·d)的劑量連續腹腔註射治療6d,于缺血後第7 d、14天分彆觀察銀杏葉組和對照組大鼠腦梗死體積、腦梗死竈週圍血管密度(層粘連蛋白,Laminin的錶達)和神經功能缺失評分的變化。結果與對照組比較,大鼠跼竈性腦缺血後第7天和14天銀杏葉組的梗死體積明顯減小,梗死週圍血管密度較對照組明顯增加,併顯示齣更好的神經功能評分(P<0.05)。結論銀杏葉提取物可促進跼竈性腦缺血後大鼠腦梗死週圍區的血管新生,從而開髮腦儲備能力,這可能是銀杏葉提取物改善腦缺血後神經功能恢複的機製之一。
목적:탐토은행협제취물시부가이촉진대서국조성뇌결혈후경사주위구적혈관신생,심구촉사뇌결혈후혈관신생、개발뇌저비능력적유잠력적중의약요법。방법채용선전법건립대서대뇌중동맥폐새(MCAO)모형,술후24h채용은행협주사액이10mg/(kg체중·d)적제량련속복강주사치료6d,우결혈후제7 d、14천분별관찰은행협조화대조조대서뇌경사체적、뇌경사조주위혈관밀도(층점련단백,Laminin적표체)화신경공능결실평분적변화。결과여대조조비교,대서국조성뇌결혈후제7천화14천은행협조적경사체적명현감소,경사주위혈관밀도교대조조명현증가,병현시출경호적신경공능평분(P<0.05)。결론은행협제취물가촉진국조성뇌결혈후대서뇌경사주위구적혈관신생,종이개발뇌저비능력,저가능시은행협제취물개선뇌결혈후신경공능회복적궤제지일。
Objective To investigate whether treatment with ginkgo biloba extract enhance angiogenesis after focal cerebral ischemia in rats. Methods Wistar rats were subjected to middle cerebral artery occlusion by intraluminal filament method. Ginkgo biloba extract (10 mg/kg) were given intraperitoneally daily for 6 consecutive days starting 24 h after ischemia. Size of infarction, vascular density in the peri-infarction region and score of neurologic function were examined on 7 d and 14 d after ischemia. Results Compared with control group at 7 d and 14 d after MCAO, infarction size of Ginkgo biloba extraxt group was sighificantly reduced. Meanwhile, vascular density in the peri-infarction region and score of neurologic function were obviously improved in the ginkgo biloba extraxt group (P<0.05). Conclusion Ginkgo biloba extract may improve angiogenesis after focal ischemia, which may be one of the neuroprotection mechanism of ginkgo biloba extract.
