目的探讨细胞增殖标记物微小染色体支持蛋白2(MCM2)在结直肠腺瘤中的表达及MCM2的表达差异与不同临床病理特征的关系。方法应用免疫组化SP法检测MCM2在结直肠腺瘤中的表达部位;应用实时荧光定量PCR法检测MCM2 mRNA在112例结直肠腺瘤中表达量的差异,同时用REST-XL?软件分析不同临床病理特征的结肠腺瘤之间MCM2的表达差异及其意义。结果MCM2在结直肠腺瘤呈全层上皮表达,MCM2 mRNA表达与结直肠腺瘤患者的年龄和腺瘤直径、形态及组织病理类型无关,不同腺瘤间不典型增生程度与MCM2 mRNA表达量有显著差异性(P <0.05)。结论细胞增殖标记物MCM2在结直肠腺瘤表达与腺瘤不典型增生程度相关,可能作为早期筛查诊断结肠癌及评估腺瘤突变的指标之一。

Objective To quantitatively compare the difference of minichromosome maintenance protein 2(MCM2) mRNA expression in colorectal adenomas with different clinicopatholigic characters. The aim was to further investigate the potential role of MCM2 in early diagnosis. Methods One hundred and twelve colorectal adenomas were collected, and each specimen was divided into two parts, one for immunohistochemistry and the other for real-time RT-PCR. Differences of MCM2 mRNA expression in colorectal adenomas with different clinicopatholigic characters were evaluated by REST-XL?. Results The expression of MCM2 mRNA was examined throughout epithelium in colorectal adenomas, and the expression of MCM2 mRNA in adenomas with high grade dysplasia was significantly higher than that in adenomas with low grade dysplasia(P =0.032). MCM2 mRNA expression elevated in adenoma with villous type, larger size, sessile morphology and older age of patients, but the significant differences were not obtained by REST-XL?. Conclusion The expression of MCM2 in colorectal adenomas relative with grade dysplasia indicate the potential value of MCM2 in early diagnosis of colonic cancer.
