研究酵母中海藻糖的提取条件,用三氯乙酸、乙醇水溶液和水作为提取溶剂提取酵母中的海藻糖,还采用了超声波辅助的水提取方法。结果为用三氯乙酸提取湿酵母中海藻糖,前3次提取率总和为96.2%;用超声波辅助提取湿酵母中海藻糖,第一次的提取率为95.1%,接近三氯乙酸的前3次提取结果,前3次提取率总和为99.1%。得到乙醇水溶液的最佳提取条件为:1 g湿酵母中加入55%的乙醇溶液9 mL,提取时间90 min,该条件下的两次提取率为81.9%。得到水提取的最佳条件为:1 g干酵母中加入12 mL水,提取温度90℃,提取时间75 min,该条件下的两次提取率为86.6%。这些提取方法比较表明,采用水提取酒香酵母中的海藻糖较经济实用。

@@@@The conditions of extracting trehalose from yeast were studied in the paper. extraction methods of trehalose from yeast with trichloroacetic acid, aqueous solution of ethanol, water as an extraction solvent were studied, water extraction method with ultrasound-assisted was also studied. The extraction rate of trehalose from wet yeast was 96.2%three times by trichloroacetic acid. The first extraction rate of trehalose from wet yeast was 95.1%by water extraction method with ultrasonic-assisted extraction , the result close to that of trichloroacetic acid extraction three times, the sum of extraction rate of trehalose three times was 99.1%. The conditions of extracting trehalose from wet yeast were studied with ethanol solution , the best extraction conditions were as follows, 9 mL 55%ethanol aqueous solution were added to 1 g wet yeast, extracting time was 90 min, the sum of twice extraction rate was 81.9%. The conditions of extracting trehalose from dry yeast were studied with water extraction method, the best extraction conditions were as follows, 12 mL water were added to 1 g dry yeast, extracting time was 75 min, extracting temperature was 90 ℃, the sum of twice extraction rate was 86.6%. The comparison of these extraction methods show that water extraction method is more economical and practical.
