目的采用HPLC法测定醋酸地塞米松搽剂中醋酸地塞米松的含量测定并进行方法学考察。方法用十八烷基硅烷键合硅胶为填充剂,甲醇-水(70∶30)为流动相,检测波长为240nm,柱温:25℃,流速:1.0mL/min,理论板数按醋酸地塞米松峰计算为7423,醋酸地塞米松峰和其它物质的分离度均>1.5。结果醋酸地塞米松的检测量为10~80μg范围内时呈良好线性关系,标准曲线:y=0.3075x+0.0318(r=0.9999),平均回收率为:99.00%,RSD=0.47%。结论该方法快速、简便、可靠、准确度高、重复性好,处方中其他成分对测定亦无干扰,分离效果好,可用于醋酸地塞米松搽剂的含量测定。

Objective?Using HPLC for the determination of dexamethasone acetate linimentum in determination of the content of dexamethasone acetate and method validation. Method Using C18 as filter, methanol-water(70∶30) as the mobile phase. detection wavelength at 240nm, column temperature:25℃,flow rate: 1.0mL/min, the number of theoretical plates by dexamethasone acetate peak was calculated as 7423, the resolution of dexamethasone acetate peak and other substances are greater than 1.5. Results It is a good linear relationship when Dexamethasone Acetate detection capacity range is 10~80μg, the standard curve:y=0.3075x+0.0318(r=0.9999),the average recovery rate was 99.00%,RSD=0.47%. Conclusion The method is rapid, simple, reliable, high accuracy, good repeatability, the other components in the prescription has no interference on the determination, and effective separation,which can be used for content determination of dexamethasone acetate linimentum.
