目的:研究乌司他丁对急性重症胰腺炎(SAP)大鼠NF-κB活性、NF-κB mRNA表达的影响。方法大鼠64只作为研究对象,随机分为对照组、S A P模型组、乌司他丁静脉注射组和乌司他丁腹腔灌注组各16只。在给药后24 h后处死大鼠,取出胰腺组织标本。对胰腺组织标本进行病理评分;测定胰腺细胞N F-κB活性及胰腺组织中NF-κB mRNA表达。比较4组大鼠病理评分;淀粉酶、脂肪酶检测结果;胰腺细胞NF-κB活性、NF-κB mRNA表达。通过各项指标判断乌司他丁及其不同给药途径对实验性急性重症胰腺炎大鼠NF-κB活性、NF-κB mRNA的影响。结果对照组与SAP模型组比较,NF-κB活性与急性胰腺炎呈正相关关系(r=0.572,P<0.05)。乌司他丁静脉注射组和腹腔灌注组对急性重症胰腺炎状况均有改善,且乌司他丁腹腔灌注组各方面均有优势,差异有统计学意义(均 P<0.05)。结论乌司他丁对急性重症胰腺炎(SAP)大鼠NF-κB活性、NF-κB mRNA表达具有明显的抑制作用,且腹腔灌注的给药途径作用更为显著。

목적:연구오사타정대급성중증이선염(SAP)대서NF-κB활성、NF-κB mRNA표체적영향。방법대서64지작위연구대상,수궤분위대조조、S A P모형조、오사타정정맥주사조화오사타정복강관주조각16지。재급약후24 h후처사대서,취출이선조직표본。대이선조직표본진행병리평분;측정이선세포N F-κB활성급이선조직중NF-κB mRNA표체。비교4조대서병리평분;정분매、지방매검측결과;이선세포NF-κB활성、NF-κB mRNA표체。통과각항지표판단오사타정급기불동급약도경대실험성급성중증이선염대서NF-κB활성、NF-κB mRNA적영향。결과대조조여SAP모형조비교,NF-κB활성여급성이선염정정상관관계(r=0.572,P<0.05)。오사타정정맥주사조화복강관주조대급성중증이선염상황균유개선,차오사타정복강관주조각방면균유우세,차이유통계학의의(균 P<0.05)。결론오사타정대급성중증이선염(SAP)대서NF-κB활성、NF-κB mRNA표체구유명현적억제작용,차복강관주적급약도경작용경위현저。
Objective To study the effect of Ulinastatin on activity of NF-κB and expression of NF-kB mRNA in rats with severe acute pancreatitis (SAP) .Methods 64 rats as the research object ,were randomly divided into the control group ,SAP model group ,ulinastatin intravenous injection group and ulinastatin peritoneal perfusion group with 16 rats in each group .The rats were killed 24 hours after administration medicines ,and the pancreatic tissue specimens were removed .Pathological score was given to record pancreas tissues ,the expression of NF-κB mRNA in pancreatic cell and NF-κB activity in pancreatic tissue were tested .The pathological score ,the detection results of am-ylase ,lipase ,expression of pancreatic cell NF-κB activity ,NF-κB mRNA of four groups of rats were compared to de-termine the effect of ulinastatin and different routes of administration on rat NF-κB ,NF-κB activity of mRNA in ex-perimental severe acute pancreatitis .Results Through the analysis of the control group and SAP model group ,NF-κB activity and acute pancreatitis were of positive correlation (r=0 .572 ,P<0 .05) .The status of severe acute pancreati-tis of the ulinastatin intravenous injection group and intraperitoneal perfusion group were improved ,and ulinastatin peritoneal perfusion group had advantages(P<0 .05) ,with statistical significance .Conclusion Ulinastatin can signif-icantly inhibit the N F-κ B ,F-κ B activity and expression of NF-κB mRNA of rats with severe acute pancreatitis (SAP) ,and intraperitoneal route of administration is more effective .
