通过不同的样品处理方式、IPG缓冲液浓度、染色方法和pH等条件的对比试验,优化建立了中华鳖肌肉组织蛋白质的双向电泳技术体系.结果表明:蛋白样品经试剂盒纯化后可显著提高等电聚焦效果; IPG缓冲液浓度为0.5%的聚焦效果好于2.0%浓度;考染所获蛋白质点较少,而银染所获蛋白质点明显增多;pH 4~7胶条的等电聚焦效果明显好于pH 3~10的胶条.

A 2-DE related technique is introduced and optimized for muscle proteome of Chinese soft-shelled turtle Pelodiscus sinensis. The comparative tests are conducted between different sample treatment, concen-tration of IPG buffer, staining methods and pH of IPG strips. The result indicate that the effect of isoelectric focusing (IEF) of purified protein sample is better than that of initial extracting protein, and that the concentration of 0.5%IPG buffer is more suitable than the concentration of 2.0%. Based on stained comparison, we suggest that the silver staining can detect more protein spots than coomassie brilliant blue (CCB) staining.
